摘 要:目的:分离并鉴定中药华山参95%乙醇提取物的乙酸乙酯和正丁醇溶性部位的化学成分。方法:采用硅胶、十八烷基硅烷键合硅胶和葡聚糖凝胶等柱色谱方法,以及半制备型高效液相色谱法等分离手段对华山参根95%乙醇提取物的乙酸乙酯和正丁醇溶性部位进行分离,通过核磁共振、质谱法等技术对所得化合物进行结构鉴定,采用1,1-二苯基-2-三硝基苯肼(DPPH)自由基模型对所得化合物的抗氧化活性进行评价。结果:从华山参正丁醇部位中分离得到3个化合物,分别鉴定为法筚枝苷(1)、苄基-O-α-L-吡喃阿拉伯糖基-(1→6)-β-D-吡喃葡萄糖苷(2)、阿魏酸(3);从乙酸乙酯部位中分离得到9个化合物,分别鉴定为异香草酸(4)、顺式阿魏酸(5)、3-吲哚甲酸(6)、阿魏酰胺(7)、反式阿魏酸酰对羟基苯乙胺(8)、N-反式-对-香豆酰基章鱼胺(9)、3-(4-羟基-3-甲氧基苯基)-N-[2-(4-羟基苯基)-2-甲氧基乙基]丙烯酰胺(10)、lactariolideⅠ(11)、coriolide(12);化合物3、5和化合物7~10质量浓度为0.3 mg·mL^(-1)时对DPPH自由基有一定的清除作用。结论:化合物1~12为首次从该植物中分离得到,化合物3表现出较强的抗氧化能力。Objective:To isolate and identify the chemical components of the ethyl acetate and n-butanol fractions of the 95%ethanol extract from the dried roots of Physochlaina infundibularis Kuang in the Solanaceae family.Methods:The ethyl acetate fraction and n-butanol fraction of the 95%ethanol extract from the dried roots of P.infundibularis Kuang were separated by silica gel,octadecly silane(ODS),Sephadex LH-20 gel column chromatography,and semi-preparative highperformance liquid chromatography(HPLC).The obtained compounds were structurally identified using techniques such as nuclear magnetic resonance(NMR)and mass spectrometry(MS).The antioxidant activity of the obtained compounds was evaluated using 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radical model.Results:Three compounds were isolated from the n-butanol fraction of P.infundibularis Kuang and identified as fabiatrin(1),benzyl-O-α-L-pyran arabinose-(1→6)-β-D-glucopyranoside(2),and ferulic acid(3).Nine compounds were isolated from the ethyl acetate fraction and identified as isovanillic acid(4),cis-ferulic acid(5),3-indolecarboxylic acid(6),ferulamide(7),trans-ferulate p-hydroxyphenylethylamine(8),N-trans-p-coumatoyloctopamine(9),3-(4-hydroxy-3-methoxyphenyl)-N-[2-(4-hydroxyphenyl)-2-methoxyethyl]acrylamide(10),lactariolideⅠ(11),and coriolide(12).Compounds 3,5,and 7-10 showed certain scavenging effects on DPPH free radicals at a concentration of 0.3 mg·mL^(-1).Conclusion:Compounds 1-12 were obtained for the first time from this plant and compound 3 showed a strong antioxidant capacity.
