摘  要:为了准确鉴定采自黑龙江省大庆市的三种牛虻,采用形态学和分子生物学两种方法进行鉴定。形态学方法主要依据形态特征,如头部基胛和触角的形状;背板和翅膀的花纹等。分子生物学方法依据线粒体细胞色素氧化酶I序列(COX 1),首次扩增这三种牛虻的COX 1序列并分析。结果表明:形态学鉴定结果三种牛虻分别为雁虻(Tabanus pleskei)、广斑虻(Chrysops vanderwulpi)和土耳其麻虻(Haematopota turkestanica)。分子生物学结果显示,三种牛虻COX 1序列的长度分别为1 506 bp、1 500 bp和1 506 bp。序列比对结果分别与分与Tabanus chrysurus(NC 062705.1)的同源性最高为96.5%、与Chrysops niger(NC 067608.1)的同源性最高为90.1%、与Haematopota pluvialis(MT 584146.1)的同源性最高为91.1%。进化分析显示分别与相应的种属在同一个进化分子上。说明三种牛虻分别为雁虻、广斑虻和土耳其麻虻,为进一步虻携带病原的研究奠定基础。Morphological and molecular biology methods were used to accurately identify three species of gadfly collected from Daqing city,Heilongjiang province.Morphological method was mainly based on morphological characteristics,the three species were Tabanus pleskei,Chrysops vanderwulpi and Haematopota turkestanica.While molecular biology method was based on mitochondrial cytochrome oxidase I sequence(COX 1).COX 1 sequences of three species were amplified for the first time,and the analysis results showed that the length of COX 1 sequence was 1506 bp,1500 bp and 1506 bp,respectively.The homology was 96.5%with Tabanus chrysurus(NC 062705.1),90.1%with Chrysops niger(NC 067608.1)and 91.1%with Haematopota pluvialis(MT 584146.1),respectively.Evolutionary analysis showed that each was on the same evolutionary molecule as the corresponding species,which explained that the three kinds of gadflies were Tabanus pleskei,Chrysops vanderwulpi and Haematopota turkestanica respectively.It laid the foundation for further research on the pathogen carrying of gadflies.
