摘 要:聚乳酸羟基乙酸共聚物(PLGA)是近年来较为热门的高分子材料,因其具有较强的成膜成囊性能及良好的生物相容性,在药物载体领域受到人们的广泛研究。为了获得理想的微球载体,对PLGA纳米微球制备的条件进行了优化。微球制备采用双溶剂乳化法,通过改变纳米微球制备过程中使用的乳化剂浓度、超声功率和超声时间等变量,对制备的纳米微球的平均粒径、多分散性指数(PDI)和zeta电位进行表征,从而确定了纳米微球制备的优化条件。结果表明,使用2%的乳化剂浓度,以60 W的超声功率进行4 min的超声,可以获得大小均一且形态良好的纳米微球,为纳米微球作为药物载体治疗各疾病的后续应用提供了重要的参考价值。Polylactic acid glycolic acid copolymer(PLGA)was a popular polymer material in recent years,because of its strong film-forming performance and good biocompatibility,had been widely studied in the field of drug carrier.In order to obtain the ideal microsphere carrier,the PLGA nano-microspheres preparation conditions were optimized.The microspheres were prepared by double solvent emulsification method,and the average particle size,polymer dispersity index(PDI)and zeta potential of the prepared nanospheres were characterized by changing the emulsifier concentration,ultrasonic power and time used in the preparation process of the nano-microspheres,thus,the optimal conditions for the nano-microspheres preparation were determined.The results showed that nano-microspheres with uniform size and good morphology could be obtained by using 2%emulsifier concentration and 60 W ultrasonic power for 4 min,which would provide an important reference value for the application of nano-microspheres as drug carrier in the treatment of various diseases.
