摘  要:独脚金内酯(strigolactones,SLs)是一种新型植物激素,参与调节植物分蘖、根系发育等多种生物学过程。通过外源施加SLs类似物GR24,探究其对两种黄芪幼苗根系发育的影响。结果表明:外源施加GR24黄芪幼苗侧根数量减少,且蒙古黄芪(AM)幼苗侧根对GR24较膜荚黄芪(AG)更敏感;在0.1和0.5μmol·L^(-1)GR24浓度处理下,促进了黄芪幼苗主根和根径的生长,根尖分生区长度和细胞数量增加,根横切面细胞数量增加,中柱直径变粗。相较于对照,根系IAA含量降低,SLs、细胞分裂素和油菜素内酯含量升高。外源施加0.5μmol·L^(-1)GR24时,细胞周期相关基因AmCYCB1;1、AmPCNA、AmCDKB和AmCYCD2;1表达均上调。综合分析,0.1μmol·L^(-1)GR24处理AM幼苗,有助于其主根的生长,减少侧根的数量,而0.5μmol·L^(-1)GR24处理AG幼苗效果较好。研究结果初步阐明了外源GR24能够抑制黄芪幼苗侧根的生长,促进主根的生长发育,为黄芪幼苗质量控制提供理论依据。Strigolactones(SLs)are new phytohormones that participate in regulating many biological processes such as root development and branching.The effects of SLs analogue GR24 on the root development of two species of Astragalus,A.membranaceus var.mongholicus(AM)and A.membranaceus(AG)seedlings were studied.The results showed that the number of lateral roots in Astragalus seedlings treated with GR24 decreased to some extent,and the lateral roots of AM seedlings were more sensitive to exogenous GR24 than those of AG seedlings.The treatment of 0.1 and 0.5µmol·L^(-1)GR24 promoted the growth of the main root and root diameter of Astragalus seedlings,increased the length and number of cells in the root tip meristem,increased the number of cells in the root cross section,and thickened the diameter of the middle column.The content of IAA in roots decreased,while the content of SLs,cytokinin and brassinosteroid increased.When 0.5µmol·L^(-1)GR24 was applied externally,the cell cycle related gene AmCYCB1;1,AmPCNA,AmCDKB and AmCYCD2;1 expression was up-regulated.According to the comprehensive analysis,0.1µmol·L^(-1)GR24 treatment of AM seedlings is conducive to the growth of its main roots and reduces the number of lateral roots,while 0.5µmol·L^(-1)GR24 treatment of AG seedlings is better.The results showed that exogenous GR24 could inhibit the growth of lateral root and promote the growth and development of taproot of Astragalus seedlings,providing a theoretical basis for the quality control of Astragalus.
