摘  要:Gretchen Hagen 3 (GH3)基因家族是一类生长素早期响应基因,在植物的生长发育及抗逆响应中发挥重要作用。本研究从凤梨草莓(Fragaria×ananassa)和森林草莓(F. vesca)基因组中分别鉴定出24个FaGH3基因和9个FvGH3基因。FaGH3基因分布于20条染色体上;FvGH3基因分布于6条染色体上。FaGH3基因大小差异较FvGH3大,但其二级结构相似。FaGH3主要分布于细胞质和细胞核, FvGH3主要分布于细胞质、细胞核和叶绿体。除了FaGH3.22为碱性蛋白,其余凤梨草莓和森林草莓GH3蛋白均为酸性亲水性蛋白。系统进化分析发现24个FaGH3和9个FvGH3可以分为3个亚家族,其中FvGH3.4和FvGH3.6为森林草莓中唯一的共线性基因,而凤梨草莓中存在大量片段重复和个别串联重复;凤梨草莓和森林草莓GH3基因成员之间主要存在正选择效应且无显著的密码子偏好性。顺式作用元件分析发现凤梨草莓和森林草莓GH3基因主要包含光响应元件、激素响应元件、胁迫响应元件及生长发育相关元件。qRTPCR结果分析表明, FaGH3和FvGH3基因在干旱胁迫下主要进行上调表达,在盐胁迫下主要进行下调表达,而在IAA处理下既存在上调表达,也存在下调表达。综上所述,凤梨草莓GH3基因家族的基因大小差异较森林草莓的大,其保守性相对较弱;FaGH3基因和FvGH3基因对PEG、NaCl和IAA处理均存在不同程度地响应。Gretchen Hagen 3(GH3)gene family is a class of auxin primary-response genes,which plays important roles in plant growth,development and stress responses.In this paper,24 FaGH3 genes and 9 FvGH3 genes were identified from the genomes of pineapple strawberry(Fragaria×ananassa)and woodland strawberry(F.vesca),respectively.The FaGH3 genes were distributed on 20 chromosomes and the FvGH3 genes were distributed on 6 chromosomes.The gene size difference of FaGH3 genes is larger than that of FvGH3,but their protein secondary structures are similar.FaGH3 proteins mainly distributed in cytoplasm and nucleus,while FvGH3 mainly distributed in cytoplasm,nucleus and chloroplast.In addition,except FaGH3.22,all the other proteins encoded by FvGH3 and FaGH3 were acid hydrophilic proteins.Phylogenetic analysis revealed that 24 FaGH3 and 9 FvGH3 could be divided into three subfamilies,among which FvGH3.4 and FvGH3.6 were the only collinear genes in woodland strawberry,while there were a large number of fragment repeats and some tandem repeats in pineapple strawberry.Moreover,there were mainly positive selection effects and no significant codon usage bias between the GH3 gene members of pineapple strawberry and woodland strawberry.Cis-acting elements analysis found that GH3 genes of woodland strawberry and pineapple strawberry mainly contained light response elements,hormone response elements,stress response elements,growth and development related elements.qRT-PCR analysis showed that FaGH3 and FvGH3 genes were mainly up-regulated under drought stress and down-regulated under salt stress,while there were both up-regulated and down-regulated expression under IAA treatment.In conclusion,the GH3 gene family of pineapple strawberry had a large difference in gene size than that of woodland strawberry,and its conservation was relatively weak.FaGH3 and FvGH3 genes responded to PEG,NaCl and IAA treatments to varying degrees.
