摘 要:目的研究心肌细胞肥厚时STAT-3与Beclin-1表达水平改变的情况,探索STAT-3和Beclin-1表达异常在心肌肥厚的发生、发展中的意义。方法心肌肥大模型的建立:实验组用异丙肾上腺素(isoproternol,ISO)(10μmol/L)和去氧肾上腺素(phenylephrine,PE)(100μmol/L)分别干预H9c2心肌细胞48 h;对照组不加药,定量聚合酶链反应(polymerase chain reaction,PCR)检测MYH7基因的表达,细胞免疫荧光染色后,利用图像软件计算细胞大小。细胞实验验证:实验组用ISO(10μmol/L)与PE(100μmol/L)分别处理H9c2心肌细胞48 h;对照组不加药,定量PCR检测STAT-3与Beclin-1的基因表达水平,Western blot检测STAT-3与Beclin-1的蛋白表达水平。结果经ISO与PE处理的H9c2心肌细胞的MYH7基因表达水平升高(P<0.005),细胞表面积增大(P<0.001)。ISO干预组STAT-3、Beclin-1的基因表达水平及蛋白水平均比对照组显著升高,差异有统计学意义(P<0.05)。PE干预组的STAT-3、Beclin-1的基因表达水平及蛋白表达水平均比对照组显著升高,差异有统计学意义(P<0.01)。结论本实验发现经ISO或PE处理所致的肥厚心肌细胞中,STAT-3与Beclin-1的基因表达和蛋白表达水平显著升高,提示STAT-3和Beclin-1的表达异常参与了心肌肥厚发生、发展,这一结果为寻找心肌肥厚治疗新靶点提供一定的线索。Objectives To observe the changes in the expressions of STAT-3 and Beclin-1 in hypertrophic cardiomyo⁃cytes and to evaluate the role of STAT-3 and Beclin-1 in the occurrence and development of myocardial hypertrophy.Methods Modeling hypertrophic cardiomyopathy:H9c2 cardiomyocytes in experimental group were treated with isopro⁃ternol(ISO)(10μmol/L)and phenylephrine(PE)(100μmol/L)for 48 h,while those in control group were not treated with drugs.The expression of MYH7 gene was detected by quantitative polymerase chain reaction(PCR).After immuno⁃fluorescence staining,the cell size was calculated by image software.H9c2 cardiomyocytes in experimental group were treated with ISO(10μmol/L)and PE(100μmol/L)for 48 h,while those in control group were not treated with drugs.The gene expressions of STAT-3 and Beclin-1 were detected by quantitative polymerase chain reaction(PCR),and the protein expressions of STAT-3 and Beclin-1 were detected by Western-blot.Results The expression of MYH7 gene was higher in H9c2 cardiomyocyte treated with ISO or PE(P<0.005),and the cell surface area of it was bigger(P<0.001).The expressions of STAT-3 and Beclin-1 genes and proteins in ISO intervention group were higher than those in control group(P<0.05).The expressions of STAT-3 and Beclin-1 genes and proteins in PE intervention group were higher than those in control group(P<0.01).Conclusions In this study,the genes and proteins expressions of STAT-3 and Beclin-1 significantly increase in hypertrophic cardiomyocytes treated with ISO or PE,suggesting that STAT-3 and Beclin-1 are involved in the occurrence and development of myocardial hypertrophy.This result provides the clue for exploring new therapeutic targets of myocardial hypertrophy.
