摘 要:【目的】考察艾草黄酮大孔树脂静态和动态的吸附和解吸效果,探究其作为天然抗氧化剂的应用潜力。【方法】通过静态和动态的吸附和解吸试验,从18种大孔吸附树脂中筛选合适的材料并进行纯化研究,采用超高效液相-高分辨质谱对其组分进行分析鉴定,并测定艾草黄酮经树脂纯化后的抗氧化效果。【结果】对18种大孔树脂的吸附和解吸性能分析表明,HPD722型大孔吸附树脂的性能最佳。以该大孔树脂作为艾草黄酮的纯化树脂,在上样液艾草黄酮粗提液质量浓度1.5 g·L^(-1)、pH值3、吸附温度25℃、流速2.0 mL·min^(-1)、上样液与树脂体积质量比V(艾草黄酮上样液/mL)∶m(大孔树脂质量/g)为10∶1,以及洗脱液为体积分数60%乙醇、pH值6、流速1.5 mL·min^(-1)、洗脱液与树脂体积质量比V(体积分数60%乙醇/mL)∶m(大孔树脂质量/g)为6∶1的条件下,艾草黄酮纯度为92.13%。超高效液相-高分辨质谱联用扫描结果共鉴定出艾草黄酮纯化物含有芦丁、槲皮素、木犀草素、高车前素和异泽兰黄素5种黄酮类化合物,同时还存在3,4-二羟基苯甲醛、3-羟基香豆素和咖啡酸3种化合物。艾草黄酮纯化物在质量浓度为1.5 g·L^(-1)时可完全清除1,1-二苯基-2-三硝基苯肼(DPPH)和羟基自由基,与维生素C的清除效果相同。【结论】HPD722大孔吸附树脂对艾草黄酮的静态和动态的吸附和解吸效果最好,纯化后的艾草黄酮纯度高,抗氧化活性好,具有作为天然抗氧化剂的应用潜力。【Objective】To investigate the static and dynamic adsorption and desorption effects of Artemisia argyi flavone on macroporous resin,and to explore its potential application as a natural antioxidant.【Method】The suitable resins were screened and purified from 18 kinds of macroporous adsorption resins by static adsorption and dynamic adsorption tests.The components of Artemisia argyi flavone purified were analyzed and identified by ultra performance liquid-high resolution mass spectrometry,and the antioxidant effect of Artemisia argyi flavone purified by resin was determined.【Results】The adsorption and desorption properties of 18 kinds of macroporous resin were evaluated,and the results showed that HPD722 macroporous resin had the best performance.The HPD722 macroporous resin was used as the purification resin of Artemisia argyi flavone with the loading liquid concentration of 1.5 g·L^(-1),pH value of 3,flow rate of 2.0 mL·min^(-1),and the ratio of loading liquid volume to resin mass of 10∶1(mL∶g),the eluent of 60%ethanol,pH value of 6,flow rate of 1.5 mL·min^(-1),and the ratio of eluent volume to resin mass of 6∶1(mL∶g).Under these conditions,the purity of Artemisia argyi was 92.13%.A total of 5 flavonoids including rutin,quercetin,luteolin,tetrachlorin and isozeelanflavin were identified in the purified artemisinin by ultra performance liquid-high resolution mass spectrometry.3,4-dihydroxy benzaldehyde,3-hydroxy coumarin and caffeic acid were also found in the purified Artemisia argyi flavone.At the concentration of 1.5 g·L^(-1),the purified Artemisia argyi could completely eliminate DPPH and hydroxyl radical,producing the same eliminating effect as that of vitamin C.【Conclusion】The HPD722 macroporous adsorption resin has the best static and dynamic adsorption and desorption effects on Artemisia argyi flavone.The purified Artemisia argyi flavone has high purity,good antioxidant activity,and has the potential to be used as a natural antioxidant.
