摘 要:目的研究葛花异黄酮对大鼠脑缺血再灌注脑组织损伤的保护作用及机制。方法将Wistar大鼠随机分为假手术组、模型组、实验组和阳性组。假手术组只分离双侧颈总动脉不进行结扎缺血;模型组、实验组和阳性组进行结扎缺血;实验组提前7天预防性灌胃给药160 mg·kg^(-1)葛花异黄酮;阳性组提前7天预防性灌胃给药10 mg·kg^(-1)依达拉奉。用脑缺血-再灌注模型对葛花异黄酮抗缺血性脑卒中活性进行评价,测定脑组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、髓过氧化物酶(MPO)、乳酸脱氢酶(LDH)活性,并通过蛋白质印迹法检测血红素氧化酶1(HO-1)、B细胞淋巴瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)蛋白表达,从抗氧化应激与凋亡方面探讨葛花异黄酮对大鼠缺血性脑卒中的保护作用。结果假手术组、模型组、实验组和阳性组的神经功能评分分别为0、(8.30±1.09)、(5.40±2.11)、(7.20±2.01)分;脑组织含水量分别为(78.50±3.60)%、(85.40±2.60)%、(77.50±2.40)%、(79.30±2.25)%;脑梗死体积分别为0、(27.60±1.60)%、(23.90±1.40)%、(24.30±1.40)%;SOD活性分别为(40.05±13.34)、(35.90±3.34)、(43.91±11.70)、(53.82±14.45)U·mg^(-1);MDA含量分别为(1.56±0.07)、(3.80±0.14)、(2.96±0.19)、(2.44±0.07)nmol·mg^(-1);MPO含量分别为(0.53±0.03)、(0.98±0.04)、(0.78±0.04)、(0.80±0.02)U·g^(-1);LDH含量分别为(99.63±7.12)、(161.63±9.82)、(149.93±3.18)、(137.54±5.33)U·L^(-1)。模型组与假手术组比较,实验组与模型组比较,差异均有统计学意义(均P<0.05)。结论葛花异黄酮能够有效改善脑缺血-再灌注大鼠神经功能障碍,其可能通过抑制氧化应激和凋亡发挥抗缺血性脑卒中保护作用。Objective To investigate the protective effect and mechanism of Pueraria lobata isoflavones on cerebral ischemia-reperfusion injury in rats.Methods Wistar rats were randomly divided into sham operation group,model group,experimental group and positive group.In sham operation group,bilateral common carotid arteries were isolated without ligation ischemia,model group,experimental group and positive group were given ligation ischemia.The experimental group was given 160 mg·kg^(-1)Pueraria lobata isoflavones 7 days in advance,and the positive group was given 10 mg·kg^(-1)edaravone 7 days in advance.The anti-ischemic stroke activity of Pueraria lobata isoflavones was evaluated by cerebral ischemia-reperfusion model.The activities of superoxide dismutase(SOD),malondialdehyde(MDA),myeloperoxidase(MPO)and lactate dehydrogenase(LDH)in brain tissue were measured,and the protein expressions of heme oxygenase 1(HO-1),B-cell lymphoma/lewkmia-2(Bcl-2)and Bcl-2 associated X protein(Bax)were detected by Western blotting.To explore the protective effect of Pueraria lobata isoflavones in rats with ischemic stroke on the aspects of antioxidant stress and apoptosis.Results The neurological function scores of sham operation group,model group,experimental group and positive group were 0,8.30±1.09,5.40±2.11,7.20±2.01;the brain water content were(78.50±3.60)%,(85.40±2.60)%,(77.50±2.40)%,(79.30±2.25%);the volume of cerebral infarction were 0,(27.60±1.60)%,(23.90±1.40)%,(24.30±1.40)%;the SOD activity were(40.05±13.34),(35.90±3.34),(43.91±11.70),(53.82±14.45)U·mg^(-1);MDA content were(1.56±0.07),(3.80±0.14),(2.96±0.19),(2.44±0.07)nmol·mg^(-1);the MPO contents were(0.53±0.03),(0.98±0.04),(0.78±0.04),(0.80±0.02)U·g^(-1);the LDH contents were(99.63±7.12),(161.63±9.82),(149.93±3.18),(137.54±5.33)U·L^(-1).There were significant differences between model group and sham operation group,between experimental group and model group(all P<0.05).Conclusion Pueraria isoflavones can effectively improve the neurological dys
