摘  要:目的观察电针“足三里”“梁门”“三阴交”对糖尿病胃轻瘫(diabetic gastro paresis,DGP)大鼠胃肠动力、肠道菌群结构及特定菌种含量的影响。方法42只SD大鼠雌雄各半随机分为空白对照组、模型组、胃复安组、电针组。按55 mg/kg剂量,采用一次性腹腔注射由柠檬酸-柠檬酸钠配制成2%浓度0.1 mmol/L链脲佐菌素结合高脂高糖不规律饮食制备DGP模型。胃复安组以1 mL/100 gL的剂量予1.7%浓度的胃复安药液灌胃,电针组取同侧“足三里”“梁门”“三阴交”电针治疗。三诺血糖仪测量大鼠血糖、酚红灌胃测量大鼠胃排空率及小肠推进率,16S rRNA高通量测序技术检测肠道菌群结构多样性,实时荧光定量PCR技术测定肠球菌、乳酸杆菌、双歧杆菌的含量。结果造模前,各组血糖值差异无统计学意义(P>0.05)。造模后,与空白对照组相比,模型组血糖值显著上升(P<0.01),胃排空率与小肠推进率显著降低(P<0.01),β多样性显示空间距离较远,各组间门水平相对丰度和特定菌种数量差异无统计学意义(P>0.05);电针组和胃复安组血糖值显著上升(P<0.01),胃排空率与小肠推进率显著降低(P<0.01),β多样性显示空间距离较近,各组间门水平相对丰度和特定菌种数量差异无统计学意义(P>0.05)。与模型组相比,电针组和胃复安组血糖值显著下降(P<0.05),胃排空率与小肠推进率显著升高(P<0.05),β多样性显示空间距离较远,各组间门水平相对丰度和特定菌种数量差异无统计学意义(P>0.05);与胃复安组相比,电针组血糖值、胃排空率与小肠推进率均差异无统计学意义(P>0.05),β多样性空间距离较近,各组间门水平相对丰度和特定菌种数量差异无统计学意义(P>0.05)。结论电针“足三里”等穴能降低DGP大鼠血糖水平、提升DGP大鼠胃肠推进率,可能与部分调控肠道菌群的结构有关。Objective To observe the effects of electroacupuncture at“Zusanli”(ST36),“Liangmen”(ST21)and“Sanyinjiao”(SP6)on gastrointestinal motility,intestinal microflora structure and specific bacterial species quantity in diabetic gastroparesis(DGP)rats.Methods Forty-two SD rats were randomly divided into blank control group,model group,metoclopramide group and electroacupuncture group.The DGP model was prepared using a one-time intraperitoneal injection of citrate-sodium citrate at a 2%concentration of 0.1 mmol/L streptozotocin combined with hyperlipidemia and high glucose irregular diet at a dose of 55 mg/kg.The metoclopramide group was infused with 1.7%concentration of metoclopramide solution at a dose of 1 mL/100 g,and the electroacupuncture group selected“Zusanli"(ST36)”,“Liangmen”(ST21)and“Sanyinjiao”(SP6)on the same side for electroacupuncture.The blood glucose of rats was measured by Sano Glucometer,the gastric emptying rate and the intestinal propulsion rate of rats were measured by gavage with phenol red,the structural diversity of intestinal flora was detected by 16S rRNA high-throughput sequencing technology,and the contents of enterococcus,lactobacillus and bifidobacterium were determined by real-time fluorescence quantitative PCR technology.Results Before modeling,there was no significant difference in blood glucose in each group.After modeling,compared with that of the blank control group,the blood glucose level in the model group was significantly increased(P<0.01).The gastric emptyrate and small intestinal thrust rate were significantly decreased(P<0.01).Theβdiversity showed a long spatial distance,and there was no statistical significance in relative abundance of phylum level and number of specific bacteria among the groups(P>0.05).The blood glucose level in the electroacupuncture group and the metoclopramide group was significantly increased(P<0.01),and the gastric empties rate and small intestinal thrust rate were significantly decreased(P<0.01).Theβdiversity showed a close sp
