远志外泌体样纳米颗粒的分离与成分鉴定

摘　　要：目的进行远志外泌体样纳米颗粒制备分析,为远志在神经系统疾病防治方面的特殊作用基础提供研究支持。方法基于超速离心法制备分离远志外泌体样纳米颗粒,经纳米粒度及Zeta电位仪和透射电镜鉴定表征,十二烷基硫酸钠-聚丙烯酰胺凝胶电泳测定蛋白分子量分布,用HPLC-MS分析鉴定外泌体样纳米颗粒的成分。结果分离得到的外泌体样纳米颗粒呈茶托状且膜结构明显,平均粒径为153.6 nm,Zeta电位为-9.2 mV;BCA法测得蛋白浓度为1.22 mg/mL,PRELNs蛋白的分子量在10~170 kDa;HPLC-MS鉴定出PRELNs中31个成分。结论PRELNs含有Tenuifoliose A、Onjisaponin B、polygalasaponinⅩⅩⅩⅡ等成分可能是其活性作用的物质基础。Objective The preparation and analysis of exosome-like nanoparticles of Yuanzh(i Polygalae Radix)(PRELNs)were carried out to provide research support for the special role of Yuanzhi(Polygalae Radix)in the prevention and treatment of nervous system diseases.Methods In this experiment,PRELNs were extracted by ultrahigh-speed centrifugation.The characterizations of PRELNs were judged by transmission electron microscopy and Zetasizer Nano ZS.The molecular weight distributions of proteins were measured via sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE).High performance liquid chromatography-mass spectrometry(HPLC-MS)was used to analyze the chemical constituents from PRELNs.Results PRELNs were saucer-like vesicles with a particle size of 153.6 nm and a Zeta potential of-9.2 mV.The protein concentration measured by BCA method was 1.22 mg/mL and the molecular weight distributions of proteins varied from 10 kDa to 170 kDa.A total of 31 ingredients were characterized from the PRELNs.Conclusion The results indicated that PRELNs contain tenuifoliose A,onjisaponin B,polygalasaponinⅩⅩⅩⅡand other ingredients,which may be the basis of its active functions.
