摘  要:目的对不同产地和不同炮制次数黄精提取物多糖含量和结构进行分析,比较不同产地和不同炮制次数黄精提取物对小鼠的体内抗氧化活性的影响。方法以不同产地和不同炮制次数黄精为原料,采用热水辅助超声提取黄精多糖、硫酸-蒽酮法测定多糖含量、离子色谱法(ion Chromatography,IC)测定单糖组成、高效凝胶色谱法(high performance gel permeation chromatography,HPGPC)测定多糖分子量分布、傅里叶红外光谱法(fourier transform infrared spectrometry,FTIR)测定官能团信息;将实验小鼠随机分为4组:一蒸一晒九华黄精组、一蒸一晒四川黄精组、九蒸九晒九华黄精组和对照组,分别给予灌胃干预,测定各组小鼠肝脾指数、采用酶联免疫法测定血清中总抗氧化能力(total antioxidant capacity,T-AOC)、总超氧化物歧化酶(total superoxide dismutase,T-SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、丙二醛(malondialdehyde,MDA)活性。结果表明一蒸一晒九华黄精提取物多糖含量最高,不同产地和炮制次数黄精提取物多糖单糖组成和分子量存在差异;红外光谱分析表明3种黄精提取物均含有β-D-吡喃糖环;体内抗氧化实验表明黄精提取物对小鼠肝脾指数没有显著影响,一蒸一晒九华黄精能够显著提高小鼠体内总抗氧化活性,并且3种黄精提取物均能够提高机体抗氧化酶活性。结论不同产地和不同炮制次数黄精多糖含量和结构存在差异,九华黄精提取物在体内的抗氧化活性略优于四川黄精。Objective The contents and structure of polygonatum polysaccharides from different producing areas and different processing times were analyzed.To compare the effects of different producing areas and different processing times of Huangjing(Polygonati Rhizoma)on the antioxidant activities of mice.Methods Different producing areas and different processing times of Huangjing(Polygonati Rhizoma)were used as raw materials.Polygonatum polysaccharide was extracted by hot water assisted ultrasound,polysaccharide content was determined by sulfuric acid-anthracone method,monosaccharide composition was determined by ion chromatography(IC).High performance gel permeation chromatography(HPGPC)and fourier transform infrared spectrometry(FTIR)were used to determine the molecular weight distribution and functional groups of polysaccharides.Experimental mice were randomly divided into four groups:The mice in one steaming and one tanning Jiuhua Huangjing(Polygonati Rhizoma)group,one steaming and one tanning Sichuan Huangjing(Polygonati Rhizoma)group,nine steaming and nine tanning Jiuhua Huangjing(Polygonati Rhizoma)group and control group were given gavage intervention respectively.Liver and spleen index of mice in each group were determined.Total antioxidant capacity(T-AOC),total superoxide dismutase,(T-SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)activities in serum were determined by enzyme-linked immunosorbent assay.Results The results showed that polygonatum polysaccharide content of the one steaming and one tanning Jiuhua Huangjing(Polygonati Rhizoma)was the highest.The monosaccharide composition and molecular weight of the polygonatum polysaccharide were different among different producing areas and processing times,and that the three extracts polygonatum containedβ-D-pyranose ring.The antioxidant test in vivo showed that the extracts had no significant effect on the liver and spleen index of mice.One steaming and one tanning Jiuhua Huangjing(Polygonati Rhizoma)could significantly improve the total antioxidan
