摘 要:目的 基于肠道菌群探究软肝冲剂抗四氯化碳(CCl4)所致大鼠肝纤维化的作用机制。方法 采用40%CCl_(4)(2 mL·kg^(-1))灌胃的方法建立大鼠肝纤维化模型,软肝冲剂干预后采用Masson染色评价抗纤维化的作用;采用16S rDNA检测技术对各组大鼠肠内容物的菌群进行检测,采用QIIME2 2019.4生物数据分析系统对样本肠道菌群Alpha多样性、Beta多样性、物种构成及相关通路进行分析。结果 Masson染色结果显示,与模型组比较,软肝冲剂各组胶原沉积明显减少,纤维间隔减少,纤维增生带变浅;Illumina Miseq测序结果显示,软肝冲剂干预后能显著提高大鼠肠道菌群的丰富度和多样性,与模型组比较具有显著性(P<0.05),与空白组比较无显著性差异。代谢通路分析显示,软肝冲剂干预后氨基酸生物合成、脂肪酸和脂质生物合成等多条代谢途径发生变化。结论 软肝冲剂对CCl_(4)所致大鼠肝纤维化具有很好的防治作用,其作用机制可能与通过调节肠道菌群的丰度进而调节氨基酸和脂质等代谢而发挥抗肝纤维化的作用。Objective Based on the intestinal flora to explore the mechanism of Ruangan Chongji(软肝冲剂)against CCl_(4)-induced hepatic fibrosis in rats.Methods The rat hepatic fibrosis model was established by intragastric administration of 40%CCl_(4)(2 mL·kg^(-1)).After the intervention of Ruangan Chongji,Masson staining was used to evaluate the anti-fibrosis effect;16S rDNA squencing technology was used to detect the intestinal flora in rats of each group,the Alpha diversity,Beta diversity,species composition and related pathways of the intestinal flora were analyze by QIIME22019.4 biological data analysis system.Results Masson staining results showed that Ruangan Chongji can significantly reduced the collagen deposition,fibrous separation and fibrous hyperplasia when compared with the model group.The results of Illumina Miseq sequencing showed that Ruangan Chongji can significantly improve the abundance and diversity of the intestinal flora in rats when compared with the model group(P<0.05),but there was no significant difference compared with those of the blank group.Metabolic pathway analysis showed that many metabolic pathways such as amino acid biosynthesis,fatty acid and lipid biosynthesis changed after the intervention of Ruangan Chongji.Conclusion Ruangan Chongji has significant effect on hepatic fibrosis in rats induced by CCl_(4),and its mechanism of action may be related to regulation of metabolism through intestinal flora.
