摘 要:目的 比较不同品系小鼠来源胚胎在低氧条件(5%O2)及常氧条件(20%O2)下体外发育情况和妊娠结局,探讨适用于评价人类体外受精实验室质量控制的体系。方法 KM小鼠、ICR小鼠、C57BL/6J小鼠、BALB/c小鼠各20只,雌雄各半。取四种品系雌性小鼠各10只常规促排卵,取卵后每个品系小鼠均随机分为低氧组与常氧组分别进行培养,进行体外受精及胚胎移植,观察并比较两组各品系小鼠囊胚形态结构、正常受精率、体外受精第2天(D2)卵裂率、D2优质胚胎率、D3优质胚胎率、囊胚率、新鲜移植妊娠率、解冻移植妊娠率。结果 与常氧组比较,低氧组KM小鼠、ICR小鼠、C57BL/6J小鼠、BALB/c小鼠囊胚体外培养均呈中空球形,囊胚均具有更加规则的形状和更高的折光度。低氧组KM小鼠、ICR小鼠、C57BL/6J小鼠、BALB/c小鼠囊胚率、新鲜移植妊娠率、解冻移植妊娠率均高于常氧组(P均<0.05),两组四个品系小鼠正常受精率、D2卵裂率、D2优质胚胎率、D3优质胚胎率比较均无统计学意义。结论 低氧条件下胚胎体外培养可提高KM小鼠、ICR小鼠、C57BL/6J小鼠、BALB/c小鼠胚胎体外发育潜能、改善妊娠结局。Objective To compare the in vitro development and pregnancy outcome of embryos from different strains of mice under hypoxia(5%O2)and normoxia(20%O2)for exploring a suitable system for evaluating the quality control of human in vitro fertilization laboratory.Methods Twenty KM mice,20 ICR mice,20 C57BL/6J mice,and 20 BALB/c mice,with half males and half females,were selected.Forty adult female mice with 10 mice in each strain received routine ovulation promotion.After ovulation,mice of each strain were randomly divided into the hypoxia group and normoxia group for culture;in vitro fertilization and embryo transfer were conducted.Blastocyst morphology,normal fertilization rate,cleavage rate of in vitro fertilization on day 2(D2),D2 high-quality embryo rate,D3 high-quality embryo rate,blastocyst rate,fresh pregnancy rate,and frozen-thawed transfer pregnancy rate of mice of each strain in the two groups were observed and compared.Results Compared with the normoxia group,the blastocysts of KM mice,ICR mice,C57BL/6J mice and BALB/c mice in the hypoxia group were all hollow and spherical,and had more regular shape and higher refractive power.The blastocyst formation rate,fresh transfer pregnancy rate and frozen-thawed transfer pregnancy rate of four strains in the hypoxia group were significantly higher than those in the normoxia group(all P<0.05).There were no significant differences in normal fertilization rate,D2 cleavage rate,D2 high-quality embryo rate,or D3 high-quality embryo rate of four strains of mice between hypoxia group and normoxia group(all P>0.05).Conclusion Embryo in vitro culture under hypoxic condition can enhance the in vitro developmental potential of embryos from KM mice,ICR mice,C57BL/6J mice,and BALB/c mice,and improve pregnancy outcomes.
