菸花苷对快速上浮脱险致减压病的预防作用

摘  要:目的研究菸花苷对快速上浮脱险致减压病大鼠的预防作用和大鼠肺组织炎性反应的影响。方法将157只实验大鼠随机分为常压对照组(n=10)、减压病组(n=30)、菸花苷高剂量组(n=40)、菸花苷中剂量组(n=40)和菸花苷低剂量组(n=37),在实验开始前30 min于尾静脉分别注射0.9%氯化钠溶液和菸花苷注射液,除常压对照组外,其余4组模拟快速上浮脱险进行减压病造模。观察5组大鼠的行为学、死亡率、肺组织湿干比、肺组织病理。常压对照组、减压病组和菸花苷高剂量组各随机选取10只大鼠检测肺组织肿瘤坏死因子-α(TNF-α)、白介素1β(IL-1β)、IL-6、细胞间黏附因子-1(ICAM-1)的含量和mRNA水平变化。结果与减压病组比较,菸花苷高、中、低3个剂量大鼠死亡率下降(P<0.05),肺水肿程度减轻,高、中剂量组大鼠行为学改善,肺组织病理改变明显好转。与减压病模型组比较,菸花苷高剂量组TNF-α、IL-1β、IL-6和ICAM-1含量显著降低(P<0.05);菸花苷组TNF-α、IL-6和ICAM-1的mRNA水平明显降低(P<0.05)。结论菸花苷对降低快速上浮脱险导致的大鼠减压病具有预防作用,这种作用可能是通过降低肺组织炎性因子水平,减轻内皮细胞损伤,保护肺组织等作用实现的。Objective To study the preventive effect of Nicotiflorin on rats with decompression sickness induced by fast buoyancy ascent escape and its influence on lung inflammation.Methods A total of 157 experimental rats were randomly divided into 5 groups:normal pressure control group(n=10),decompression sickness group(n=30),high-dose Nicotiflorin group(n=40),medium-dose Nicotiflorin group(n=40)and low-dose Nicotiflorin group(n=37).Nicotiflorin was injected into the tail vein of the latter three groups 30min prior to the modeling,and the same volume of 0.9%sodium chloride solution was injected into the tail vein of rats in the decompression sickness group.Except for those of the normal pressure control group,rats in the remaining groups were subjected to the fast buoyancy ascent escape to induce decompression sickness.The behavior testing,mortality,lung wet-to-dry ratio and lung pathology of rats were observed.Ten rats were randomly selected from normal pressure control group,decompression sickness group and high-dose Nicotiflorin group to detect the contents and mRNA levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),IL-6 and Intercellular adhesion molecule-1(ICAM-1).Results Compared with those of decompression sickness group,decreased mortality and alleviated pulmonary edema were detected in Nicotiflorin groups(P<0.05).Behavioral testing data and pathological lesions in the lung were significantly improved in rats of high-dose and medium-dose Nicotiflorin group(P<0.05).Compared with those of decompression sickness group,the contents of TNF-α,IL-1β,IL-6 and ICAM-1 were significantly reduced in high-dose Nicotiflorin group(P<0.05),and the mRNA levels of TNF-α,IL-6 and ICAM-1 were significantly reduced in Nicotiflorin groups(P<0.05).Conclusion Nicotiflorin presents a preventive effect on alleviating decompression sickness in rats induced by fast buoyancy ascent escape via reducing the levels of inflammatory factors in lung tissue,alleviating endothelial cell damage,and protecting lung tissues.
