大豆SVP基因进化分析及调控开花功能研究

摘  要:SVP(SHORT VEGETATIVE PHASE)基因是MADS-box家族一员,它通过介导不同开花途径调控下游基因表达,从而影响植物开花。本研究主要探究大豆SVP基因是否参与调控开花,为进一步深入解析大豆SVP基因的主要功能和调控机制提供依据。本研究利用同源比对与进化树分析鉴定出大豆SVP基因GmSVP01和GmSVP02,利用定量PCR检测GmSVP01和GmSVP02基因的表达模式以及不同生长发育时期的转录水平变化,通过拟南芥回补实验验证GmSVP01和GmSVP02基因调控开花的功能,并利用已发表的424份大豆自然群体材料的重测序数据对大豆SVP基因进行单倍型分析。结果表明:GmSVP01和GmSVP02两个基因主要在大豆营养生长阶段表达,且在顶芽和侧芽中的表达量较高,暗示其可能与组织分化及调控开花期的功能有关。长日照条件下的转基因拟南芥表型分析表明大豆SVP基因能够回补拟南芥svp突变体的早花表型,证明大豆GmSVP01和GmSVP02基因具有调控开花的功能,且与拟南芥SVP基因功能相似。单倍型分析表明GmSVP01和GmSVP02在大豆的进化过程中受到的选择程度不同。综上所述,本研究克隆了大豆两个SVP基因,初步阐明了其时空表达模式,并证明GmSVP01和GmSVP02基因与拟南芥同源基因功能相似,具有调控开花期的功能。As a member of the MADS-box family,SVP(SHORT VEGETATIVE PHASE)regulates flowering time through different pathways in Arabidopsis.But the function of GmSVP on flowering time in soybean is unclear.This study mainly explored whether the GmSVP is involved in regulating flowering,which will provide basis for further understanding the main functions and regulatory mechanisms of GmSVP in soybean.Homologous comparison and evolutionary tree analysis were used to identify GmSVP01 and GmSVP02 in soybean,then the reverse transcription and quantitative polymerase chain reaction was used to unravel the expression pattern of GmSVP01 and GmSVP02 during the different developments and on different organizations.In addition,the two genes were transformed into Arabidopsis svp mutants to verified the function on flowering time.Finally,we used the sequencing data of 424 germplasm resource to analyze the haplotype of GmSVP01 and GmSVP02.Our research indicated GmSVPs regulate flowering time,and found that GmSVPs preferentially expressed in shoot apical meristems and axillary meristems,and mainly expressed in vegetative growth than the reproductive growth.Under long-day photoperiod,the transgenic plant flowered later than svp mutant,which demonstrated that the GmSVP01 and GmSVP02 could regulate flowering and are functionally equivalent to the Arabidopsis SVP gene.The analysis on the haplotype of the SVP gene indicated that GmSVP01 and GmSVP02 undergo the different degrees of selection during the evolution.In summary,we cloned GmSVP01 and GmSVP02,clarified their spatiotemporal expression patterns,and demonstrated that GmSVP01 and GmSVP02 perform similar function to SVP in Arabidopsis on regulating flowering time.
