摘 要:NAC转录因子是植物中最大的特异性转录因子家族之一,广泛参与植物生长发育过程,在植物受到逆境胁迫应答后能够发挥重要的作用。为了解析GmNAC120基因的功能,本研究从大豆cDNA中克隆出了GmNAC120基因,采用生物信息学方法分析基因序列及其编码蛋白质的特征和结构,采用酵母单杂试验分析编码蛋白的转录激活活性。结果表明:GmNAC120基因开放阅读框为1296 bp,共编码431个氨基酸,分子量为49.684 kD,等电点为4.91。GmNAC120蛋白序列3~153氨基酸位置为1个高度保守的NAC结构域。GmNAC120蛋白不存在信号肽,也不存在跨膜结构,属于亲水性蛋白,位于细胞核中。GmNAC120与野大豆中的NAC蛋白的亲缘性关系较近,氨基酸序列一致性高达99.54%。酵母单杂交试验证明GmNAC120基因具有转录激活活性。NAC transcription factors are one of the largest families of specific transcription factors in plants.NAC transcription factors are not only widely involved in plant growth and development,but also play an important role in plant response to stress.In order to analyze the function of GmNAC120 gene,GmNAC120 gene was cloned from soybean cDNA.Bioinformatics method was used to analyze the gene sequence and the characteristics and structure of the encoded protein.The transcriptional activation activity of the encoded protein was analyzed by yeast single hybrid test.The results showed that the open reading frame of GmNAC120 gene was 1296 bp,encoding 431 amino acids,with a molecular weight of 49.684 kDa and an isoelectric point of 491.GmNAC120 contains a highly conserved NAC domain at position 3-153.There is no signal peptide and transmembrane structure in GmNAC120 protein.GmNAC120 is a hydrophilic protein and is located in the nucleus,closely related to NAC protein in wild soybean,and the amino acid sequence identity is up to 99.54%.The GmNAC120 gene was proved to be with transcriptional activation activity by yeast single hybrid test.
