摘　　要：单细胞转录组测序(single-cell RNA sequencing,scRNA-seq)技术能够揭示同一组织中异质细胞的表达谱,在细胞发育轨迹研究中发挥重要作用。目前,拟南芥(Arabidopsis thaliana)、水稻(Oryza sativa)以及玉米(Zea mays)等植物中已经逐步开展单细胞转录组研究,但相比动物细胞,植物中单细胞转录组应用尚处于起步阶段。本文阐述了植物单细胞转录组测序常用技术及其在组织发育机制、植物细胞动态发育轨迹、细胞间分化方向与互作关系等研究中的应用,讨论了单细胞转录组测序技术应用在植物中存在的问题和挑战,并对其发展前景进行展望,为利用单细胞转录组测序技术探究植物组织发育机制提供参考。Single-cell RNA sequencing(scRNA-seq)is an advanced technique for characterizing the expression profiles of heterogeneous cells in the same tissue,and plays a crucial role in the study of cell development trajectory.Compared with animal cells,the application of scRNA-seq in plants is still in its infancy,and has been gradually applied in Arabidopsis,rice(Oryza sativa)and maize(Zea mays).This review summarized the common techniques of scRNA-seq and their applications in the research of tissue development mechanism,cells dynamic development trajectory,direction of cells differentiation and interaction.Additionally,this article analyzed the problems,challenges and development prospects of the application of scRNA-seq in plants,hoping to provide a theoretical basis for explore the mechanism of plant tissue development.
