摘  要:为了预测蔗糖转化酶基因家族在蔗糖代谢中的作用,本研究利用生物信息学方法对西瓜所有蔗糖转化酶基因家族成员进行搜索鉴定及分析。结果显示:西瓜蔗糖转化酶基因家族一共包括12个基因,系统发育分析揭示,这些基因分为3个已知的亚群,其中细胞壁蔗糖转化酶基因(CWINV)5个,液泡蔗糖转化酶基因(VINV)2个,细胞质蔗糖转化酶基因(CINV)5个,它们分别分布在6条染色体上,甜瓜的蔗糖转化酶基因数量及类型与西瓜相同,分布在8条染色体上。细胞壁蔗糖转化酶和液泡蔗糖转化酶同属于酸性蔗糖转化酶,细胞质蔗糖转化酶属于中性/碱性蔗糖转化酶,这两者的保守基序类型不同,保守基序motif 1、motif 7在所有酸性蔗糖转化酶蛋白中是高度保守的。中性/碱性蔗糖转化酶蛋白的保守性要高于酸性蔗糖转化酶蛋白,几乎所有中性/碱性蔗糖转化酶蛋白都含有motif 2、motif 3、motif 4、motif 5、motif 9、motif 10这6个保守基序。基因结构显示,部分酸性蔗糖转化酶基因存在外显子丢失情况。西瓜和甜瓜的蔗糖转化酶基因家族上游序列存在多种顺式作用元件,包括光响应、激素应答、逆境响应、发育调节四大类。根据定量分析的结果,推测ClCWINV1、ClCWINV2、ClCWINV5、ClCINV1、ClCINV3、ClCINV4、ClCINV5可能是西瓜蔗糖代谢的关键基因。本研究的结果为进一步阐述西瓜蔗糖转化酶基因家族成员如何调控果实蔗糖代谢提供一定的理论依据。In order to predict the role of sucrose invertase gene family in sucrose metabolism in watermelon fruit,this study used bioinformatics methods to search and identify all the sucrose invertase gene family members of watermelon,and analyzed it.The results showed that the watermelon sucrose invertase gene family includes a total of 12 genes.Phylogenetic analysis segregated the putative sucrose invertase into three distinctively known sub-groups,of which CWINV has 5,VINV has 2 and CINV has 5,and they are all distributed on 6 chromosomes.The number and type of sucrose invertase genes in melon were the same as those in watermelon,which were distributed on 8 chromosomes.Cell wall sucrose invertase and vacuolar sucrose invertase belong to acid sucrose invertase,cytoplasmic sucrose invertase belongs to neutral/alkaline sucrose invertase,and the two types of conservative motifs are different.motif 1,motif 7 is highly conserved among all acid sucrose invertase proteins.The conservation of neutral/alkaline sucrose invertase protein is higher than that of acid sucrose invertase protein.Almost all neutral/alkaline sucrose invertase proteins contain 6 conserved motifs:motif 2,motif 3,motif 4,motif 5,motif 9,and motif 10.The gene structure suggests that some acid sucrose invertase genes have exon loss.There is a variety of cis-regulatory elements in the upstream sequence of the watermelon and melon sucrose invertase gene family,including four categories:light response,hormone response,abiotic stress response,and developmental regulation.According to the results of quantitative analysis,it is speculated that ClCWINV1,ClCWINV2,ClCWINV5,ClCINV1,ClCINV3,ClCINV4,ClCINV5 may be key genes for watermelon sucrose metabolism.The results of this study serves as a theoretical basis for further elucidating how members of the sucrose invertase gene family regulate sucrose metabolism in watermelon fruit.
