滇一型粳稻雄性不育系细胞质基因组的溯源分析

摘　　要：滇一型和BT型是杂交粳稻生产应用中仅有的两类不育系,到目前为止,对滇一型粳稻雄性不育细胞质的来源并不清楚。为明确滇一型粳稻细胞质雄性不育基因的来源,本研究从滇一型不育系中克隆了细胞质雄性不育基因orf79D1,发现它与BT型不育系中orf79基因序列完全一致;诱导ORF79D1蛋白在大肠杆菌中表达,发现其对细菌的生长繁殖有明显的抑制作用。利用我们开发的滇一型细胞质雄性不育基因特异的功能分子标记,对来源于云南不同地域的53份云南地方籼稻种质资源的分析发现,有22份资源具有滇一型细胞质雄性不育基因,经测序分析发现22份种质中的orf79D1基因序列完全一致。说明具有orf79D1不育基因的细胞质广泛存在于云南的地方籼稻种质资源中,滇一型粳稻雄性不育细胞质应来源于云南地方籼稻种质。本研究为进一步探究滇一型粳稻雄性不育细胞质特性奠定了基础。Dian-typel(Dl)and BT types are the only two types of sterile lines used in the production application of keng hybrid rice.So far,the origin of the male sterile cytoplasm of D1 CMS is not clear.To determine the origin of the cytoplasmic male sterility gene of D1 keng rice,This research the cytoplasmic male sterility gene orf79D1 was cloned from the Dian-1 sterile lines and found to be identical to the sequence of orf79 gene in the BT sterile lines;The ORF79D1 protein was induced to be expressed in Escherichia coli and found to have a significant inhibitory effect on the growth and reproduction of the bacteria;The analysis of 53 Yunnan local hsien rice germplasm resources from different regions of Yunnan showed that 22 resources had the Dian-1 cytoplasmic male sterility gene using our developed Dian-1 cytoplasmic male sterility gene-specific functional molecular markers.Sequencing was analyzed found that resulted in complete sequence identity of the orf79D1 gene in 22 germplasm.This indicated that cytoplasm with orf79D1 sterility gene is widely present in local hsien rice germplasm resources in Yunnan,and D1 keng rice male sterile cytoplasm should be derived from Yunnan local hsien rice germplasm.In this study,the basis was laid for further investigation of the characteristics of D1 keng rice male sterile cytoplasm.
