油菜硫苷近红外光谱分析模型的建立

摘  要:为探索甘蓝型油菜硫代葡萄糖苷近红外分析模型建立的方法,本研究选取了291份甘蓝型油菜材料,利用氯化钯比色法测定油菜籽的硫苷含量,采用近红外光谱分析仪The Unscrambler软件系统对数据进行分析和处理,最终建立了近红外光谱模型。其中,氯化钯比色法条件优化为提取时间50 min,室温(24℃)下显色时间为2 h,在波长540 nm处测定吸光度值。回收率在95.55%~105.62%之间,平均回收率为100.47%,精密度试验标准偏差为0.95μmol/g,精密度较高,重复性好,可用于油菜籽硫苷含量的测定,也可用于油菜硫苷近红外光谱分析模型的建立。近红外分析模型的硫苷定标决定系数(RSQ)与交叉验证决定系数(1-VR)都超过了0.85(越接近1越理想),校正标准差(SEC)和校正内部检验标准差(SECV)均小于0.4(越小越理想)说明模型回归曲线与实测点拟合度高,预模型测性好,可用于油菜品种的筛选。试验优化了氯化钯比色法测油菜籽硫苷含量的条件,建立的近红外模型可准确、快速、无污染、低消耗地测定油菜籽硫苷含量。To explore the method of near infrared analysis model of glucosinolates in Brassica napus L.,in this study,291 rapeseed materials were selected.The content of glucosinolate in rapeseed was determined by palladium chloride colorimetric method,and the data were analyzed and processed by The Unscrambler software system with near infrared spectroscopy.Finally,a near infrared spectral model was established.The optimized conditions of palladium chloride colorimetric method were as follows:extraction time of 50 min,color development time of 2 h at room temperature(24 ℃),and absorbance value at 540 nm.The recoveries were between 95.55%and 105.62%,the average recoveries were 100.47%,the standard deviation of precision test was 0.95μmol/g,the accuracy was high,the reproducible was good,and it could be used for the determination of rapeseed glucosin content,and the establishment of rapeseed glucosin analysis model by near infrared spectroscopy.Near infrared analysis model of glucosinolates scaling determination coefficient(RSQ)and cross validation decision coefficient(1-VR)were more than 0.85(the more close to 1,the ideal),and standard deviation correction(SEC)and the internal inspection standard deviation correction(SECV)were less than 0.4(ideal),the smaller model fit of the regression curve and the measured point was high,the forecast model of measurement and can be used for the screening of rapeseed varieties.The experiment optimized the conditions for the determination of glucosinolate content in rapeseed by palladium chloride colorimetric method.The near-infrared model established can be used to determine glucosinolate content in rapeseed accurately,rapidly,without pollution and with low consumption.
