响应面法优化川西獐牙菜中三种环烯醚萜苷的提取工艺研究

摘　　要：对川西獐牙菜中三种环烯醚萜苷抗炎活性成分进行提取工艺研究,建立快速高效的提取方法,提高药材的利用率。以川西獐牙菜中獐牙菜苦苷、龙胆苦苷、獐牙菜苷三种环烯醚萜苷为研究对象,考察提取次数、液料比、提取时间、乙醇浓度以及提取温度5个因素对川西獐牙菜中三种环烯醚萜苷提取率的影响,通过响应面法和Box-Behnken优化得到最优工艺参数。结果表明:川西獐牙菜中三种环烯醚萜苷的最优提取工艺参数为:提取次数2次,提取时间60 min,液料比46 mL·g^(-1),乙醇浓度73%,提取温度50℃;验证结果显示在该条件下川西獐牙菜中三种环烯醚萜苷的提取率为4.73 mg·g^(-1),与预测值4.75 mg·g^(-1)相差0.0255 mg·g^(-1),RSD<5%。实验所采用的超声法提取川西獐牙菜中三种环烯醚萜苷的优化工艺可行,耗时短、效率高、简单易行,能够满足提取要求。The extraction technology of three iridoid glycosides from Swertia mussoti Franch.was studied to provide reference data for the pharmacological and pharmacodynamic studies and utilization of the active components of S.mussoti.,to improve the efficiency of resources usage,and offer reference basis for the rational exploitation and utilization of S.mussotii..The effects of extraction times,solid-liquid ratio,ethanol concentration,extraction time and extraction temperature on the extraction amount of iridoid glycosides in S.mussoti.were investigated.Response surface methodology(RSC)and Box-Behnken were used to optimize the extraction process parameters.The results show that The optimal extraction parameters of the three iridoid glycosides from S.mussoti were as follows:liquid-solid ratio 46 mL·g^(-1),ethanol concentration 73.37%,extraction temperature 50°C,extraction time 60 min,extraction times 2times.The results showed that the extractionamount of the three iridoid glycosides in S.mussoti.was 4.73 mg·g^(-1) under these conditions,which was 0.0255 mg·g^(-1) different from the predicted value of 4.75 mg·g^(-1),with RSD<5%.The method of ultrasonic extraction of the three iridoid glycosides from S.mussoti.is feasible,with short time consuming,high efficiency and simple operation,which can meet the extraction requirements.
