摘 要:探索病毒致病机制有助于靶向药物和疫苗研发,对疫病防控具有重要意义。单颗粒示踪(single particle tracking,SPT)技术能够在活细胞中对单个病毒粒子进行实时成像,便于观察病毒与宿主细胞相互作用的动态过程,以揭示病毒的入侵机制,因而成为病毒机制研究的重要工具。本文简要介绍了SPT技术的发展和技术原理,重点阐述了SPT技术在猪伪狂犬病病毒、日本脑炎病毒、猪繁殖与呼吸综合征病毒、猪流行性腹泻病毒和猪传染性胃肠炎病毒的入胞、胞内转运机制研究中的应用,旨在为相关领域研究提供参考。The research on pathogenic mechanism of viruses is conducive to the development of targeted drugs and vaccines,which is of great significance for disease prevention and control.Real-time imaging for single virion could be conducted within live cells by single-particle tracking(SPT)technology,which is convenient to observe the dynamic process of the interaction between viruses and host cells,and thus the invasion mechanism of viruses is revealed.Therefore,SPT could be used as a key tool for the research on mechanism.For the purpose of providing a reference for future research on relevant fields,the development and technical principle of SPT was briefly introduced,especially its application in the research on entry of cell and intracellular transport mechanisms of porcine pseudorabies virus(PRV),Japanese encephalitis virus(JEV),porcine reproductive and respiratory syndrome virus(PRRSV),porcine epidemic diarrhea virus(PEDV)and porcine transmissible gastroenteritis virus(TGEV).
