摘 要:分析贝母药材中的生物碱类化学成分与药理活性。择取A549、NCI-H157、16HBE、SK-MES-1及NCI-H460非小细胞肺癌细胞株资料进行研究,通过UHPLC-QTOF-MS/MS(超高效液相色谱仪与四极杆飞行时间串联质谱仪)结合细胞代谢组学技术观察贝母药材中的生物碱类化学成分对肿瘤细胞增殖的影响,分析其药理活性。进一步明确贝母对不同种类非小细胞肺癌细胞株的抗肿瘤作用。结果表明:A549和NCI-H157的侵袭转移能力越强,16HBE及NCI-H460次之,而贝母药材中的生物碱类化学成分的抗肿瘤药理活性突出。贝母提取物的药理活性与药物提取浓度呈正相关,生物碱类化学成分提取率越高,药物活性成分越强。Objective:To analyze the chemical constituents and pharmacological activities of alkaloids in Fritillaria.Method:Non-small cell lung cancer cell lines A549,NCI-H157,16HBE,SK-MES-1 and NCI-H460 were selected for study.The effects of alkaloids in Fritillaria fritillaria on the proliferation of tumor cells were observed by UHPLC-QTOF-MS/MS(ultra-high performance liquid chromatograph and quadrupole time-of-flight tandem mass spectrometer)combined with cell metabolomics technology,and their pharmacological activities were analyzed.To further clarify the antitumor effect of Fritillaria fritillaria on different types of non-small cell lung cancer cell lines.The results:The invasion and metastasis ability of A549 and NCI-H157 was stronger,followed by 16HBE and NCI-H460,and the alkaloid chemical components of Fritillaria had prominent anti-tumor pharmacological activity.Conclusion:The pharmacological activity of Fritillaria fritillaria extract is positively correlated with the concentration of drug extraction.The higher the extraction rate of alkaloid chemical components,the stronger the drug active components.
