硝矾洗剂对急性炎症的作用及其机制研究

摘　　要：目的:研究硝矾洗剂抗炎的作用机制,验证其临床用量(原液稀释1~2倍)的合理性。方法:采用醋酸致小鼠扭体、二甲苯致小鼠耳肿胀、角叉菜胶致大鼠足肿胀3种急性炎症模型,以10%洁尔阴洗液为阳性组,评价硝矾洗剂低(原液稀释两倍)、中(原液稀释一倍)、高(原液)浓度组的抗炎作用,比较各组行为学差异及血清中环磷酸腺苷(cAMP)、前列腺素E2(PGE2)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)的含量。结果:与模型组相比,阳性组和硝矾洗剂各浓度组均能有效减少小鼠扭体次数、小鼠耳肿胀程度和大鼠足肿胀程度,也能降低老鼠血清中cAMP、PGE2、IL-1β、TNF-α的含量。特别地,硝矾洗剂发挥抗炎作用呈浓度相关性,低浓度组即有良好的效果,中、高浓度组的效果优于阳性组,且中、高浓度组差异无统计学意义。结论:硝矾洗剂具有良好的抗炎效果,其作用机制可能与血清中cAMP、PGE2、IL-1β、TNF-α的含量下降有关,也表明了硝矾洗剂临床用量的合理性。Objective:To preliminarily study the mechanism of the anti-inflammatory action of nitrate alum lotion and confirm the rationality of its clinical dosage(diluted 1~2 times of the stock solution).Methods:Writhing in mice induced by acetic acid,ear swelling in mice induced by xylene,and foot swelling in rats induced by carrageenan were used as acute inflammation models.The anti-inflammatory effects of the high(stock solution),medium(diluted once of the stock solution),and low(diluted twice of the stock solution)concentration groups were compared to the positive group(1o percent Jieeryin lotion),with behavioral differences and serum levels of cyclic adenosine monophosphate(cAMP),prostaglandin E2(PGE2),interleukin-1β(IL-1p)and tumor necrosis factor-αlpha(TNF-α)being assessed.Results:The positive group and each concentration group of nitrate alum lotion can effectively reduce the number of writhing in mice,the degree of ear swelling in mice,and the degree of swelling in rats'feet,as well as serum levels of cAMP,PGE2,IL-1β,and TNF-αcontent when compared to the model group.In particular,the anti-inflammatory effect of nitrate alum lotion is concentration-dependent,the low concentration groups have good effects,the effect of the medium and high concentration groups is better than that of the positive group,and there is no significant difference between the medium and high concentration groups.Conclusion:Nitrate alum lotion has a good anti-inflammatory effect,and its mechanism of action may be related to the decrease of the contents of cAMP,PGE2,IL-1β,and TNF-αin serum.In addition,it also shows the rationality of the clinical dosage of nitrate alum lotion.
