摘 要:目的:比较摘花或摘果处理对牡丹皮品质的影响。方法:以浸出物得率、丹皮酚含量及HPLC指纹图谱测得的特征化学成分为评价指标,比较各试验组样本品质差异。浸出物测定采用药典方法,丹皮酚含量检测及HPLC指纹图谱的色谱条件:采用C18色谱柱(4.6 mm×250 mm,5μm),以0.01%磷酸水溶液-乙腈为流动相,梯度洗脱,流速为1.0 mL/min,检测波长为250 nm,柱温为30℃,进样体积为10μL。结果:经摘花或摘果处理后,牡丹皮浸出物得率增高,但丹皮酚含量降低。建立的9个不同处理牡丹皮样本的指纹图谱,其相似度>0.97;标定了8个共有特征色谱峰,指认了5个特征成分(没食子酸、氧化芍药苷、芍药苷、苯甲酰芍药苷、丹皮酚)。与对照组比较,摘花组和摘果组8个共有特征色谱峰峰面积均有6个降少、2个增加。结论:摘花或摘果处理对牡丹皮化学成分的作用结果,因成分不同而不同,该处理可能不利于牡丹皮药用品质提升。Objective:To compare the effects of flower-picking or fruit-picking on the quality of Moutan Cortec.Methods:The extraction rate,the content of paeonol and the characteristic chemical components measured by HPLC fingerprint were used as evaluation indexes to evaluate the samples quality among experimental groups.The contents of ethanol-soluble extract was deter mined by Chinese Pharmacopoeia 202o.The content deter mination of paeonol content detection and HPLC fingerprint chromatographic separation was achieved on a Cis Column(4.6mm X 250 mm,5μm),phosphoric acid aqueous solution and acetonitrile was used as mobile phase in gradient elution program with the flow rate of 1.0 mL/min,the column temperature of 30℃C,the injection volume of 50μL and the detection wavelength of 260 nm.Results:After the treatment of flower or fruit picking,the extract yield of moutan increased,but the paeonol content decreased.The fingerprint of 9 moutan samples with different treatments were established,which contained the 8 common peaks,and 5 chromatographic peaks were identified,namely:gallic acid,oxypaeoniflorin,paeoniflorin,benzoyl paeoniflorin,paeonol.The similarity of each fingerprint is higher than O.97.Compared with the control group,the peak areas of the 8 common feature peaks in the lower-picking and fruit-picking flower groups decreased in 6 and increased in 2.Conclusion:The effects of flower-picking or fruit-picking on chemical constituents of Moutan Cortex were vary from component to componen,and this method may not be conducive to the quality of Moutan Corte.
