摘  要:目的:考察蒙药藁本水蒸气蒸馏法(SD法)与超临界CO_(2)萃取法(SFE-CO_(2)法)提取部位化学成分的差异,为进一步研究蒙药藁本化学成分及其药理活性提供依据。方法:采用SD法与SFE-CO_(2)法分别提取蒙药藁本挥发油;通过稀释、过滤制备供试液;气相色谱-质谱联用技术(GC-MS)进行定性分析;色谱峰面积归一法计算各组分的相对含量。结果:共鉴定出30个成分;其中,SD法提取部位鉴定出19个成分,峰面积占总峰面积的62.33%,主要成分是(E)-3-丁烯基-4,5-二氢-1(3H)-异苯并呋喃酮,相对含量是SFE-CO_(2)部位的两倍多;SFE-CO_(2)法提取部位鉴定出17个成分,峰面积占总峰面积的53.97%,主要成分是8,11-十八碳二烯酸甲酯,SD部位未检测到;共有成分有6个,共有成分在SD部位与SFE-CO_(2)部位中的相对含量不同;两种提取部位的化学成分种类及相对含量也有差异。结论:SD部位的主要成分为藁本内酯(3-丁烯基-4,5-二氢-1(3H)-异苯并呋喃酮),与文献报道一致,且SD部位检测出的化学成分总相对含量高于SFE-CO_(2)部位。研究不同提取部位化学成分的区别,为进一步研究蒙药藁本化学成分及更好开发利用藁本提供实验依据。Objective:To compare the chemical components of extracts by steam distillation(SD)and supercritical CO2 fluid extraction(SFE-CO_(2))from mongolian medicine Ligustici Rhizoma.The results provided basis for further study on chemical components and pharmacological activities of mongolian medicine Ligustici Rhizoma.Methods:SD and SFE-CO_(2)methods were used to extract the essential oil from mongolian medicine Ligustici Rhizoma;The test solution was prepared by dilution and filtration;Qualitative analysis was performed by gas chromatography-mass spectrometry technique(GC-MS);The relative contents of each component were calculated by peak area normalization method.Results:A total of 30 components were identified,of which 19 components were identified by SD and the peak area accounted for 62.33%of the total peak area,the main component is(E)-3-butylidene-4,5-dihydroisobenzofuran-1(3H)-one,the relative content is more than twice that of SFE-CO_(2)extract.And 17 components were identified by SFE-COz and the peak area accounted for 53.97%of the total peak area,the main component is 8,ll-octadecadienoic acid,methyl ester,the SD extract was not detected.There are 6 common components,the relative contents of common components in SD and SFE-CO_(2)extracts were different.The types and relative contents of chemical components of the two extracts were also different.Conclusion:The main component of SD extract was ligustilide(3-butylidene-4,5-dihydroisobenzofuran-1(3H)-one),which was consistent with the literature reports.The total relative content of chemical components detected in SD extract was higher than that in SFE-CO_(2)extract.The differences of chemical components of different extracts were studied in order to provide experimental basis for further study of chemical components and better development and utilization of Mongolian medicine Ligustici Rhizoma.
