摘 要:目的:构建特色院内制剂润喉饮治疗急性咽炎的“成分-靶点-通路”网络关系,预测其潜在作用机制,为院内制剂开发提供理论依据。方法:通过TCMSP、SYMMAP、ETCM、SwissTargetPrediction数据库收集润喉饮中五种中药(十大功劳、地黄、木蝴蝶、桔梗及淡竹叶)的活性成分筛选并预测其作用靶点。利用TTD、DrugBank、GeneCards数据库收集和筛选急性咽炎发生相关靶点;通过STRING数据库构建作用靶点之间交互网络,并导入Cytoscape 3.7.1软件构建“成分-靶点”网络。通过R语言对筛选后的靶点进行GO和KEGG富集分析,采用Network Analyzer分析网络参数,最终得到润喉饮治疗急性咽炎的“成分-靶点-通路”核心网络,采用分子模拟对接技术验证。结果:共筛选和收集得到润喉饮治疗急性咽炎活性化合物90个,潜在作用靶点424个。富集分析结果显示,润喉饮中的活性成分能调控IL-17、AGE-RAGE、Small cell lung cancer等信号通路,其中槲皮苷、木樨草素、黄芩素和蒲公英赛醇可能是治疗急性咽炎的主要活性成分,MAPK1、AKT、NF-κB、IKBKB、AP-1可能是潜在作用靶点。结论:润喉饮可能通过调控AKT/IKBKB/NF-κB和MAPK1/AP-1信号通路抑制炎症因子的表达而发挥抗炎作用。Objective:To construct the“ingredient-target-pathway"network relationship of our special formulation of Runhou Decoction(RHD)for the treatment of acute pharyngitis,and to predict its potential mechanism of action,so as to provide a theoretical basis for the development of the formulation in the hospital.Methods:The active ingredients of five herbal medicines in RHD(Mahoniae Eadir,Radir Rehmanniae Recen,Indian Trumpet flower Seed,Platycodon Root and Lophatherum gracile)were collected from TCMSP,SYMMAP,ETCM and SwissTargetPrediction databases and their targets were predicted.The TTD,DrugBank and GeneCards databases were used to collect and select the targets related to the development of acute pharyngitis;the STRING database was used to construct the interaction network among the targets,and Cytoscape 3.7.1 software was used to construct the"ingredient-target"network.The GO and KEGG enrichment analysis of the screened targets were performed by R language,and the parameters of the network were analyzed by Network Analyzer to obtain the core network of"ingredient-targetpathway"for the treatment of acute pharyngitis.Molecular simulation docking technique was used to verify the core active ingredients and their targets.Results:90 active ingredients and 424 potential targets were obtained.The results of enrichment analysis showed that the active ingredients in RHD can regulate IL-17,AGE-RAGE and other signaling pathways,among which quercetin,luteolin,baicalin and taraxerol may be the main active ingredients for the treatment of acute pharyngitis,and AKT,AP-1,NF-κB and MAPK1 may be the potential action targets.Conclusion:RHD inhibits the expression of inflammatory factors by regulating AKT/IKBKB/NF-κB and MAPK1/AP-1 signaling pathways.
