摘 要:目的:研究三种色胺类物质AMT、5-MeO-AMT和5-MeO-DiPT诱导的小鼠条件性位置偏爱(CPP)行为.方法:通过CPP实验方法对AMT、5-MeO-AMT和5-MeO-DiPT的奖赏效应进行评估,采用腹腔注射方式,以MDMA为阳性对照,观察不同剂量的AMT、5-MeO-AMT和5-MeO-DiPT小鼠训练5 d后获得的CPP分数.结果:CPP实验结果显示,MDMA在给药剂量为3 mg/kg时开始能够剂量依赖性地诱导小鼠产生CPP行为,AMT和5-MeO-AMT在给药剂量为30 mg/kg时能够诱导小鼠产生CPP行为,在本研究剂量范围内,5-MeO-DiPT不能诱导小鼠的CPP行为.结论:与MDMA相比,这三种色胺类物质AMT、5-MeO-AMT和5-MeO-DiPT诱导小鼠产生CPP行为的奖赏效应强度较弱,提示其精神依赖性可能较弱,其具体作用机制有待于进一步研究.Objective:To investigate the conditioned place preference(CPP)behavior induced by three tryptamines AMT,5-MeOAMT and 5-MeO-DiPT in mice.Methods:In this paper,the rewarding effects of AMT,5-MeO-AMT and 5-MeO-DiPT were tested by the CPP test.MDMA was used as the positive control.Intraperitoneal injections of AMT,5-MeO-AMT and 5-MeO-DiPT at different doses were used to observe the CPP scores of miceobtained after 5 days of training.Results:The results of the CPP test showed that MDMA started to dose-dependently induce CPP behavior in mice at the dose of 3 mg/kg,while AMT and 5-MeO-AMT started to induce CPP behavior in mice at the dose of 30 mg/kg.5-MeO-DiPT failed to induce CPP behavior in mice within the range of doses in this experiment.Conclusion:The rewarding effects of CPP behavior induced by the three tryptamines AMT,5-MeO-AMT and 5-MeO-DiPT are weaker than MDMA,suggesting that their psychological dependence may be weaker.The specific mechanism of them needs to befurther investigated.
