摘  要:采用超高效液相色谱-串联四级杆质谱联用法(HPLC-MS/MS)测定氮离子(N+)注入后金莲花(Trollius chinensis Bunge)中荭草苷(orientin)、牡荆苷(vitexin)和藜芦酸(veratric acid)含量。样品经甲醇提取后,以超纯水和乙腈为流动相,选用Agilent ZORBAX XDB-C18色谱柱洗脱分离,采用多反应监测模式(MRM)和电喷雾离子源(ESI)负离子模式检测。结果表明:荭草苷、牡荆苷、藜芦酸分别在20~500、40~500、10~1000 ng·mL^(-1)范围内线性关系良好,决定系数均大于0.99;该方法的精密性和稳定性良好,分别在2.23%~3.19%和5.30%~6.94%之间;3种化合物的平均加标回收率在97.06%~98.54%之间,相对标准偏差在2.23%~5.22%之间。该方法具有良好的灵敏度和准确性,适合金莲花中荭草苷、牡荆苷和藜芦酸含量的检测。此外,荭草苷和藜芦酸分别在N+注入剂量为5×10^(15)、2×10^(15)ions·cm^(-2)时其含量最高,牡荆苷在各N+注入剂量下含量均低对照。ULTRA-performance liquid chromatography-tandem quadrupole mass spectrometry(HPLC-MS/MS)was used to determine the content of orientin,vitexin and veratric acid in Trollius chinensis Bunge after nitrogen ion implantation.The samples were extracted with methanol and eluted on an Agilent ZORBAX XDB-C18 column using ultrapure water and acetonitrile as mobile phases.The detection was performed in multiple reaction monitoring mode(MRM)and ionization spray ion source negative ion mode(ESI).The linear range of Orientin,Vitexin and Veratric acid was respectively 20-500,40-500 and 10-1000 ng·mL^(-1)with correlation coefficients greater than 0.99.The precision and stability of this method were in the range of 2.23%~3.19%and 5.30%~6.94%respectively.The average recoveries of the three compounds ranged from 97.06%to 98.54%,and relative standard deviation ranged from 2.23%to 5.22%.This method had good sensitivity and accuracy,and was suitable for the determination of Orientin,Vitexin and Veratric acid in T.chinensis Bunge.The results showed that the contents of Orientin and Veratric acid were the highest at 5×10^(15)and 2×10^(15)ions·cm^(-2)respectively,while Vitexin contents were lower than control at any dosage of nitrogen ions.
