摘  要:目的基于网络药理学探讨葛根治疗风寒感冒的药理机制。方法通过TCMSP数据库获取葛根的有效化学成分及其靶点基因,筛选主要化学成分并建立化合物-靶点网络。通过TTD、Gencards数据库获取风寒感冒相关疾病的靶点,运用R语言获取药物与疾病的交集基因,利用STRING11.5平台对交集基因进行分析,构建PPI网络并获取潜在靶点,将PPI网络数据输入CytoScape3.9.1软件进行拓扑分析得到核心靶点。采用Metascape进行GO富集分析和KEGG分析,而后采用CytoScape3.9.1软件构建“GOBP-基因-KEGG”网络。结果通过TCMSP数据库获取葛根有效化学成分3个,葛根靶点基因398个;筛选出疾病靶点1242个;通过R语言得到葛根和风寒感冒交集的12个重要靶点,包括NOS2、PTGS1、PTGS2等;通过拓扑分析得到核心靶点5个,分别为NOS2、PTGS1、PTGS2、MAPK14、HSP90AA1;通过对葛根与风寒感冒的交集基因进行GO富集分析得到9个富集的生物过程聚类,KEGG分析得到6个富集的通路聚类。结论通过网络药理学探究了葛根治疗风寒感冒的基因靶点和富集通路。Objective To investigate the pharmacological mechanism of Pueraria lobata in the treatment of wind-chill cold based on network pharmacology.Methods The effective chemical ingredients and target genes of Pueraria lobata were obtained through TCMSP database,the key chemical ingredients were screened,and the compound-target network was established.Targets of diseases related to wind-chill cold were obtained through TTD and Gencards databases,and the intersection genes of drugs and diseases were obtained by R language.The intersection genes were analyzed by using STRING11.5 platform.PPI network was constructed,and potential targets were obtained.The PPI network data were input into CytoScape3.9.1 software for topological analysis to obtain the key targets.Metascape was used for GO enrichment analysis and KEGG analysis,and then"GOBP-Gene-KEGG"network was constructed by CytoScape3.9.1 software.Results A total of 3 effective chemical ingredients of Pueraria lobata and 398 target genes of Pueraria lobata were obtained by TCMSP database;A total of 1242 disease targets were screened out;Through R language,12 important targets of the intersection of Pueraria lobata and wind-chill cold were obtained,including NOS2,PTGS1,PTGS2,etc.;5 key targets were obtained by topological analysis,namely NOS2,PTGS1,PTGS2,M APK14 and HSP90AA1.Thro ugh GO enrichment analysis of the intersection genes of Pueraria lobata and wind-chill cold,9 enriched biological process clusters were obtained,and 6 enriched pathway clusters were obtained by KEGG analysis.Conclusion The gene target and enrichment pathway of Pueraria lobata in the treatment of wind-chill cold were investigated through network pharmacology.
