摘 要:目的 通过观察针刺对ApoE^(-/-)小鼠胆固醇代谢的影响,探讨针刺改善动脉粥样硬化(atherosclerosis,AS)的作用及可能机制。方法 选取ApoE^(-/-)小鼠制备AS模型,随机分为模型组、针刺组及阳性药物对照组,另取C57BL/6J小鼠为正常组。治疗结束后,采用HE染色观察主动脉斑块形成情况,ELISA法检测血清胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白(high-density lipoprotein cholesterol,HDL-C)及低密度脂蛋白(low-density lipoprotein cholesterol,LDL-C)水平,Western blot和荧光定量PCR法分别检测肝脏组织中腺苷三磷酸结合盒转运体G5(ATP-binding cassette subfamily G member 5,ABCG5)、ABCG8蛋白与m RNA表达水平,以及小肠组织中ABCG5、ABCG8、胆固醇调节元件结合蛋白2(Sterol-regulatory element binding protein 2,SREBP2)、尼曼-匹克C1型类蛋白(Niemann-pick C1L1,NPC1L1)的蛋白与mRNA表达水平。结果 与正常组比较,模型组小鼠动脉内膜下可见明显粥样斑块,TC、TG及LDL-C表达显著升高,HDL-C表达显著降低(P<0.01),肝脏与小肠组织ABCG5、ABCG8蛋白及mRNA表达显著降低(P<0.01),小肠组织SREBP2、NPC1L1蛋白及mRNA表达显著升高(P<0.01);与模型组比较,针刺组与阳性药物对照组小鼠动脉内膜下粥样斑块显著减少,TC、TG及LDL-C表达显著降低,HDL-C表达显著升高(P<0.01),肝脏与小肠组织ABCG5、ABCG8蛋白及mRNA表达显著升高(P<0.01),小肠组织SREBP2、NPC1L1蛋白及m RNA表达显著降低(P<0.01)。结论 针刺可能通过调节胆固醇的代谢从而改善AS。Objective By observing the effect of acupuncture on cholesterol metabolism in ApoE^(-/-)mice,to explore the effect and possible mechanism of acupuncture on atherosclerosis(AS).Methods ApoE^(-/-)mice were selected to prepare AS model and randomly divided into model group,acupuncture group and positive drug control group,and C57BL/6J mice were taken as normal group.After treatment,HE staining was used to observe the formation of aortic plaque,and ELISA was used to detect the levels of total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C),western blot and real-time fluorescence quantitative PCR were used to detect the expression of ATP binding cassette subfamily G member 5(ABCG5),ABCG8 protein and mRNA levels in liver tissues,and the expression of ABCG5,ABCG8,Sterol-regulatory element binding protein 2(SREBP2),Niemann pick type C1 protein(NPC1L1)protein and mRNA expression levels in small intestine tissues.Results Compared with the normal group,there were obvious atherosclerotic plaques under the intima of the model group,the expression of TC,TG and LDL-C increased significantly,the expression of HDL-C decreased significantly(P<0.01),the expression of ABCG5,ABCG8 protein and mRNA decreased significantly(P<0.01),and the expression of SREBP2 and NPC1L1 protein and mRNA increased significantly(P<0.01).Compared with the model group,the subintimal atherosclerotic plaque in the acupuncture group and the positive drug control group decreased significantly,the expressions of TC,TG and LDL-C decreased significantly,the expression of HDL-C increased significantly(P<0.01),the expression of ABCG5,ABCG8 protein and mRNA increased significantly(P<0.01),and the expression of SREBP2 and NPC1L1 protein and mRNA decreased significantly(P<0.01).Conclusion Acupuncture may improve AS by regulating cholesterol metabolism.
