摘 要:目的 分析患者外周血蛋白组学变化,寻找不同阶段爆震损伤患者外周血差异蛋白及相关通路。方法分别选取短期及长期爆震损伤患者作为研究对象,提取外周血血清,通过蛋白质组学DIA技术筛选出外周血显著差异表达的蛋白,并进行生物信息学分析。结果 短期损伤组相对正常组差异表达蛋白共95种,主要包括ALDOA等;生物分析显示以上蛋白主要富集于糖酵解等通路;长期损伤组相对正常组差异表达蛋白共21种,主要包括SOD3等;生物分析显示以上蛋白种类主要集中在胆固醇代谢通路。结论 爆震性耳聋发生早期可出现细胞间黏附性降低与炎症相关蛋白增加,长期接触爆震声患者的载脂蛋白表达增多。以上相关蛋白及通路的发现为爆震性聋发生后不同阶段干预提供潜在靶点。Objective To identify differentially expressed proteins and related pathways in peripheral blood in patients with blast injury hearing loss at different stages via analyzing proteomics changes.Methods Peripheral blood serum was extracted from patients with short-term or long-term blast injury hearing loss,and differentially expressed proteins were screened by proteomic DIA technology and analyzed by bioinformatics.Results Compared with normal controls,95 proteins were found to be differentially expressed in patients with short-term blast injury hearing loss,includ‐ing ALDOA.Bioanalysis showed that these proteins were mainly enriched in platelet degranulation and other pathways.In patients with long-term blast injury hearing loss,21 proteins were differentially expressed as compared with normal controls,mainly involving SOD3.Bioanalysis showed that these protein types were mainly concentrated in the cholesterol metabolism pathway.Conclusion In the early stage of blast induced deafness,intercellular adhesion may be decreased and inflammation-related proteins increased.Long-term exposure to blast noise can cause increased apolipoprotein expression.These findings provide potential targets for interventions at different stages of blast injury deafness.
