摘  要:目的:分析通络祛痛膏挥发油成分,并进行透皮试验,考察挥发油不同成分的透皮能力,以指导产品质量控制。方法:水蒸气蒸馏法提取通络祛痛膏及其单味药材挥发油,Franz扩散池法进行体外透皮试验,GC-MS联用技术获得总离子流图,各色谱峰相应的质谱图经过NIST17.L标准谱库检索进行定性分析,根据峰面积归一化法进行定量分析,计算各成分的相对含量。结果:在成品中共鉴定出49种成分,占挥发油总量的98.42%,42种成分可归属于处方原药材;透皮接受液中共鉴定出9种有效成分,D-(+)-樟脑(17.41%)、薄荷脑(6.28%)、龙脑(9.75%)、异龙脑(3.36%)、丁香酚(36.5%)、对甲氧基肉桂酸乙酯(3.14%)、水芹烯(0.17%)、金合欢烯(0.05%)及桂皮醛(0.65%)是通络挥发油的主要透皮成分。结论:通络祛痛膏挥发油成分复杂多样,但在透皮吸收后的成分明显减少,通过透皮吸收透过率定性分析其透皮效果,其中,桂皮醛的透过率最高,D-(+)-樟脑的累积透过量最高;该研究运用GC-MS技术全面分析了通络祛痛膏中挥发性成分及透皮前后化学成分的变化,可为通络祛痛膏的质量控制提供科学依据。OBJECTIVE To analyze the components of the volatile oil of Tongluo Qutong Gao and conduct the transdermal experiment to investigate the transdermal ability of different components of the volatile oil,so as to guide the quality control of the product.METHODS The volatile oil of Tongluo Qutong Gao and its single medicinal material was extracted by steam distillation.The Franz diffusion cell method was used for in vitro transdermal experiment.The total ion flow diagram was obtained by GC-MS.the corresponding mass spectra of each color spectrum peak were searched by NIST17.L standard library for qualitative analysis.The quantitative analysis was carried out according to the peak area normalization method,and the relative percentage content of each component was calculated.RESULTS A total of 49 components were identified in the finished product,accounting for 98.42%of the total volatile oil,and42 components could be attributed to the original medicinal materials.A total of 9 effective components were identified by transdermal liquid,including D-(+)-camphor(17.41%),menthol(6.28%),borneol(9.75%),isoborneol(3.36%),eugenol(36.5%),ethyl p-methoxycinnamate(3.14%),carylene(0.17%),acanthene(0.05%)and cinnamaldehyde(0.65%).CONCLUSION The components in the volatile oil of Tongluo Qutong Gao are complex and diverse,but the components are significantly reduced after transdermal absorption.The transdermal effect is qualitatively analyzed by its transdermal absorption and permeability.cinnamaldehyde has the highest permeability and D(+)-camphor has the highest cumulative permeability.In this study,GC-MS technology was used to comprehensively analyze the volatile components in Tongluo Qutong Gao and the changes of chemical components before and after transdermal,so as to provide scientific basis for the quality control of Tongluo Qutong Gao.
