摘 要:目的动物实验是现代毒理学实验的重要方法之一,半数致死量(LD_(50))作为重要的实验指标,往往需要大量的计算过程,本研究探讨应用Stata 15.1软件进行毒性实验中LD_(50)的计算,以减轻实验人员的计算负担。方法将从文献获得的急性毒性实验数据输入Stata 15.1软件中,第一步对毒物剂量以10为底数作为对数变换,将结果作为自变量;第二步以死亡动物数量r的二项分布为因变量,用probit连接函数作广义线性模型拟合,以Bliss法进行LD_(50)的计算;最后应用“delta”法对加权直线回归方程的参数进行估计,并计算标准误,进而得到LD_(50)的置信区间(CI)。结果用Stata15.1软件可同时计算出LD_(50)及其95%CI。该结果与使用SAS和SPSS软件分析得出的LD_(50)结果基本一致,差异无统计学意义(P>0.05)。结论Stata 15.1软件可用于计算LD_(50),与使用其他昂贵或大体量软件分析得出的结果一致,同时Stata 15.1软件具有简单、直观和可编程的特点,易于推广应用。Objective Animal experiments were one of the important methods in modern toxicology experiments.As an important experimental index,median lethal dose(LD_(50))often required a lot of calculation process.This study discussed the application of Stata 15.1 software to calculate LD_(50)in toxicity experiments,so as to reduce the calculation burden of laboratory personnel.Methods To input the acute toxicity experiments data obtained from the literature into Stata 15.1 software.The first step was to make a logarithmic transformation of the toxic dose based on 10 and take the result as an independent variable;in the second step,the binomial distribution of the number of dead animals r was taken as the dependen variable,the probit connection function was used to fit the generalized linear model,and the Bliss algorithm was used to calculate the LD_(50);finally,the"delta"method was used to estimate the parameters of the weighted linear regression equation,and the standard error was calculated to obtain the confidence interval(CI)of LD_(50).Results Using Stata 15.1 software,LD_(50)and 95%CI could be calculated simultaneously.The results were generally consistent with those obtained for the LD_(50)using SAS and SPSS software analysis,and the differences were not statistically significant(P<0.05).Conclusion Stata 15.1software could be used to calculate LD_(50).It was consistent with the results obtained by using other expensive or large-scale software,and Stata 15.1 sofltware had the characteristics of simplicity,intuition and programmability,and was easy to popularize and apply.
