金汁对脓毒症小鼠血清炎症因子表达的影响

摘　　要：目的观察金汁对脓毒症小鼠血清炎症因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)表达的影响。方法30只小鼠随机分为正常对照组、模型组、生理盐水组、金汁组和金汁+PDTC组,每组各6只。金汁+PDTC组造模前2 h进行腹腔注射吡咯烷二硫代氨基甲酸酯(PDTC)120 mg/kg。脓毒症模型选用小鼠经盲肠结扎穿孔法建立,造模后生理盐水组用生理盐水0.01 mL/g灌胃,金汁组、金汁+PDTC组用金汁0.01 mL/g灌胃,灌胃时间为造模后12、24、36 h,共3次。正常对照组和模型组不予任何处置。造模后48 h观察小鼠一般情况并取材,采用苏木精-伊红染色法观察金汁对脓毒症小鼠肠组织损伤的影响,采用酶联免疫吸附法检测各组小鼠血清中IL-1β、IL-6、TNF-α的表达情况。结果HE染色可见与模型组比较金汁组固有层炎症细胞明显减少。与正常对照组相比,模型组炎症因子IL-1β、IL-6和TNF-α表达均显著升高(P<0.05);而与模型组比较,金汁治疗组血清组织中IL-1β、IL-6和TNF-α表达均下降(P<0.05)。结论金汁可明显改善脓毒症小鼠的炎症损伤,对其促炎因子的表达具有一定的抑制作用,从而发挥治疗脓毒症的作用。Objective:To investigate the effect of Jinzhi on the expression of serum inflammatory factors inter-leukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in septic mice.Methods:Thirty mice were randomly grouped according to the experimental design:normal control group,sepsis model group(CLP group),sepsis model+normal saline administration group(CLP+NaCl group),sepsis model+Jinzhi administration group(CLP+JZ group)and sepsis model+Jinzhi+PDTC administration group(CLP+JZ+PDTC group),with six mice in each group.PDTC was injected intraperitoneally(120 mg/kg intraperitoneally,one injection)2 h before modeling in the CLP+JZ+PDTC group.The sepsis model was established by cecal ligation and puncture,and the CLP+NaCl group was treated with saline(0.01 mL/g)by gavage,and the CLP+JZ and CLP+JZ+PDTC groups were treated with Jinzhi(0.01 mL/g)by gavage(gavage time was 12,24 and 36 h after modeling for a total of three times),the control group and CLP group received no treatment.The general state in health of the rats was ob-served and taking material was conducted 48 h after modeling.The effect of Jinzhi on intestinal tissue injury in septic mice was observed by hematoxylin-eosin staining(HE),and the expression of IL-1β,IL-6 and TNF-αin the serum of each group was detected by enzyme-linked immunosorbent assay(ELISA).Results:HE staining showed a significant reduction of inflammatory cells in the lamina propria of CLP+JZ group compared with CLP group.The expression of inflammatory factors IL-1β,IL-6 and TNF-αwere significantly increased in the surgical group of septic mice compared with the control group(P<0.05),while the expression of IL-1β,IL-6 and TNF-αin serum tissues decreased in the Jinzhi treated group compared with the CLP group(P<0.05).Conclusion:Jin-zhi can significantly improve the inflammatory damage of septic mice and has a certain inhibitory effect on the ex-pression of pro-inflammatory factors,thus playing a role in the treatment of sepsis.
