摘　　要：目的利用网络药理学技术,探究骨痹方治疗膝骨关节炎(KOA)的作用机制。方法通过中药系统药理学数据库与分析平台(TCMSP)和中药分子机制的生物信息学分析工具(BATMAN-TCM)等数据库获取骨痹方潜在活性成分及对应靶点,通过Drugbank、GeneCards、OMIM、TTD和DigSee数据库得到KOA疾病靶点。使用String数据库构建PPI网络,采用Cytoscape 3.8.0软件筛选核心靶点和关键成分;通过Metascape数据平台对核心靶点进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)富集分析。结果筛选出骨痹方的关键成分10个,作用于白蛋白(ALB)、白细胞介素-6(IL-6)等核心靶点,通过调控晚期糖基化终产物及其受体(AGE-RAGE)等信号通路发挥作用。结论骨痹方通过多成分、多靶点、多通路发挥治疗KOA的作用。Objective To explore the potential mechanism of Gubi recipe in the treatment of knee osteoarthritis based on network pharmacology.Methods TCMSP and BATMAN-TCM database were used to mine the potential active ingredients and target of Gubi recipe.Databases such as Drugbank,Genecards,OMIM,TTD and Digsee were used to screen the targets of knee osteoarthritis.The protein protein interaction(PPI)network of target protein was constructed by String database.Core targets and key ingredients were screened by Cytoscape 3.8.0 software.Key targets were analyzed by Metascape for GO functional and KEGG signaling pathway enrichment.Results Ten key ingredients of Gubi Recipe were screened,which act on ALB,IL6 and other core targets,and play a role by regulating AGE-RAGE and other signal pathways.Conclusion Gubi Recipe played a role in the treatment of knee osteoarthritis through multi-ingredients,multi-target and multi-pathway.
