摘  要:目的:通过高通量测序及生物信息学分析方法寻找紫朱软膏干预下肢静脉性溃疡的可能机制。方法:紫朱软膏治疗下肢静脉性溃疡大白兔模型3只,应用二代测序技术寻找用药前后差异LncRNA,利用生物信息学分析方法分析差异LncRNA,初步建立下肢静脉性溃疡差异表达LncRNA与靶基因及基因功能调控网络。结果:紫朱软膏外用新西兰大白兔下肢静脉性溃疡疮面中存在176个有差异表达的LncRNA,其中上调有91个,下调有85个,确定了10个差异表达的LncRNA。mRNAs有1 801个,上调、下调的mRNAs分别有751、1 050个;GO分析结果表明,差异表达的LncRNA分别富集在生物学过程(BP)、细胞成分(CC)、分子功能(MF)。KEGG富集分析结果表明,差异表达的LncRNA主要富集在与细胞周期、肿瘤相关的通路和信号转导通路上。结论:初步建立下肢静脉性溃疡差异表达LncRNA与靶基因及基因功能调控网络。Objective:To explore the possible mechanism of Zizhu Ointment in the intervention of venous ulcer of lower limbs by high-throughput sequencing and bioinformatics analysis.Methods:Three rabbit models of venous ulcer of lower limbs were treated with Zizhu Ointment.The second generation sequencing technology was used to find the difference of LncRNA before and after treatment.The bioinformatics analysis method was used to analyze the difference of LncRNA.The differential expression of LncRNA,target gene and gene function regulation network of venous ulcer of lower limbs were preliminarily established.Results:There were 176 differentially expressed LncRNAs in venous ulcer of lower limbs of New Zealand white rabbits treated with Zizhu ointment,91 of which were up-regulated and 85 of which were down-regulated.A total of 1801 mRNAs,751 up-regulated and 1050 down-regulated mRNAs;The results of GO analysis showed that the differentially expressed mRNAs were enriched in biological process(BP),cellular component(CC)and molecular function(MF)respectively.The results of KEGG enrichment analysis showed that differentially expressed mRNAs were mainly enriched in cell cycle,tumor related pathways and signal transduction pathways.Conclusion:The network of differentially expressed LncRNA,target gene and gene function regulation in venous ulcer of lower extremities was preliminarily established.
