摘  要:目的:在药辅合一理论下,探索对白及多糖(BSP)进行氧化改性使其作为活性高分子材料应用的可能性。方法:柱前衍生化HPLC分析BSP的单糖组成,高碘酸钠法对BSP进行改性,傅里叶红外光谱、核磁共振光谱进行产物结构表征,划痕实验、MTT实验评价产物的生物活性,Schiff碱反应制备交联化水凝胶,流变仪考察交联的影响因素。结果:BSP由葡萄糖和甘露糖组成,氧化改性后获得了侧链具有活性醛基的氧化白及多糖(OBSP),pH=5条件下可与绿原酸接枝壳聚糖自发交联,形成具有组织黏附性和自愈合性的新型多糖基水凝胶,温度和氧化度是影响交联的主要因素。结论:氧化改性后的BSP是有潜力的活性高分子材料。Objective:To explore the potential of Bletilla striata polysaccharides(BSP)as bioactive polymer material after oxidation modification under the guidance of unification of medicine and adjuvant theory.Methods:Monosaccharide composition of BSP was analyzed by PMP-HPLC.BSP was oxidized by sodium periodate method.Product was identified by FTIR and NMR.The in vitro activity of product was evaluated by wound healing assay and MTT assay.Cross-linked hydrogels were prepared by Schiff base reaction.Influencing factors of crosslinking was analyzed through rheometer.Results:BSP is composed of glucose and mannose.Oxidized BSP is highly active both chemically and biologically.A novel self-healing hydrogel with tissue adhesion was formed through crosslinking between oxidized BSP and chlorogenic acid grafted chitosan under pH=5.Temperature and oxidation degree of oxidized BSP are the main factors affecting crosslinking.Conclusion:BSP after oxidation modification is a potential active polymer material.
