摘  要:目的:基于血管内皮生长因子(VEGF)/血管内皮生长因子受体2(VEGFR-2)和血管生成素(Ang-1)/酪氨酸激酶-2(Tie-2)信号通路,探讨养精种玉汤调控薄型子宫内膜(TE)大鼠子宫血管生成的机制。方法:选取动情周期规律的雌性未交配SD大鼠30只,随机分为对照组、模型组、中药组,每组10只,宫腔注射95%乙醇建立TE模型,分组给药14 d,处死后剖腹取子宫,采用HE染色观察子宫形态,WesternBlot检测大鼠子宫VEGF、VEGFR-2、Ang-1、Tie-2蛋白表达,Quantitative Real-Time PCR检测大鼠子宫VEGF、VEGFR-2、Ang-1、Tie-2mRNA表达水平。结果:与模型组比较,中药组能明显改善模型大鼠的子宫内膜形态,增加子宫湿重和子宫脏器指数(P<0.05),并接近对照组水平;中药组大鼠子宫VEGF、VEGFR-2、Ang-1、Tie-2蛋白及mRNA水平均显著增加(P<0.05,P<0.01)。结论:养精种玉汤可以明显改善TE大鼠的子宫组织形态,增加其内膜血管生成,可能与提高大鼠子宫组织VEGF、VEGFR-2、Ang-1、Tie-2表达水平有关。Objective:To investigate the mechanism of Yangjing Zhongyu Decoction regulating uterine angiogenesis in rats with thin endometrium(TE)based on the VEGF/VEGFR-2 and Ang^(-1)/Tie-2 signaling pathways.Methods:Thirty female unmated SD rats with regular estrous cycle were selected and randomly divided into control group,model group and traditional Chinese medicine group,with 10 rats in each group.The TE model was established by intrauterine injection of 95%absolute ethanol.The rats were administered into groups for 14 days,and the uterus was collected by laparotomy.HE staining was used to observe the endometrial morphology,Western Blot was used to detect the protein expressions of VEGF,VEGFR-2,Ang^(-1) and Tie-2 in the rat uterus,and Quantitative Real-Time PCR was used to detect the VEGF,VEGFR-2,Ang^(-1) and Tie-2 mRNA expression in the rat uterus.Results:Compared with the model group,the traditional Chinese medicine group could significantly improve the endometrial morphology of the model rats,increase the wet weight of the uterus and the uterine organ index(P<0.05),and were close to the level of the control group,the uterine VEGF,VEGFR-2,Ang^(-1),and Tie-2 protein and mRNA levels of the rats in the traditional Chinese medicine group were increased(P<0.05,P<0.01).Conclusion:Yangjing Zhongyu Decoction can significantly improve the uterine tissue morphology of rats with TE and increase endometrial angiogenesis,which may be related to the increase of VEGF,VEGFR-2,Ang^(-1) and Tie-2 in rat uterine tissue.
