摘  要:目的通过构建复合EGFL7植入物的去细胞神经支架移植修复大鼠坐骨神经损伤缺损模型,从而探究EGFL7在周围神经修复过程中促进血管化和神经恢复的作用。方法手术切取日本大耳白兔双侧胫神经,截成1.0 cm长的节段,制备去细胞神经支架。将制备好的外源性EGFL7的去细胞神经支架复合物植入大鼠坐骨神经损伤模型,于术后4周取标本行神经电生理检测、组织取材投射电镜形态学观察等来评价神经修复效果。结果添加外源性EGFL7模型组与不添加EGFL7的模型组相比再生的有髓神经纤维数量多、形态规则、直径粗、髓鞘厚、髓鞘不折叠或崩解、髓鞘发育良好;坐骨神经测得的诱发电位曲线波幅高、潜伏期短、神经传导速度快。结论EGFL7在周围神经修复过程中可起到促进血管化和神经功能恢复的作用。Objective To study the promotion of vascularization and neural recovery by EGFL7 in peripheral nerve repair,by constructing decellularized Neural Scaffolds witn complex EGFL7 which is used to repair rat sciatic nerve injury.Methods The dissection of the tibial nerves from the legs of Japanese white rabbits,which was to prepare decellularized neural scaffolds by cutting into 1.0 cm long segments.Implanted the prepared decellularized neural scaffold complex with exogenous EGFL7 into the rat sciatic nerve injury model,to Evaluate nerve repair effects by Nerve electrophysiological testing and projection electron microscope observation with specimens in 4 weeks after surgery.Results In the Model with the addition of exogenous EGFL7,the number of myelinated nerve fibers was large,the shape was regular,the diameter was thick,the myelin sheath was thick,the myelin sheath was not folded or disintegrated,and the myelin sheath was well developed,when compared with the model without the addition of exogenous EGFL7.Model with the addition of exogenous EGFL7 had the evoked potential curve measured by the sciatic nerve has high amplitude,short latency and fast nerve conduction velocity.Conclusion EGFL7 acts to promote vascularization and recovery of nerve function in the during peripheral nerve repair.
