摘 要:为探索紫花苜蓿(Medicago sativa L.)雄性不育的分子机制,选取紫花苜蓿细胞质雄性不育系(MSJN1A)与保持系(MSJN1B)花蕾发育早期的花药进行细胞学观察,并采用高通量Illumine Hiseq2000测序平台,进行转录组测序,对测序结果注释、筛选与分析。结果表明:花粉败育始于四分体后期,于单核早期出现明显的不育现象;两系花药间共存在4284个差异表达基因(Differentially expressed genes,DEGs)(FDR<0.001,|log 2FC|>2);GO功能注释分析表明,DEGs涉及物质代谢、细胞过程、转运蛋白活性等功能;KEGG通路富集分析表明,DEGs涉及淀粉与蔗糖代谢、戊糖与葡萄糖醛酸相互转化、糖酵解/糖异生、苯丙烷生物合成等途径;转录因子分析表明,DEGs涉及bHLH,MYB,PHD等已知与花粉发育相关的转录因子家族。本研究通过对紫花苜蓿花药结构观察和转录组分析,为研究紫花苜蓿雄性不育成因机制提供参考。To explore the molecular mechanism of male sterility in alfalfa(Medicago sativa L.),anthers from early bud development of alfalfa cytoplasmic male sterile lines(MSJN1A)and maintainer lines(MSJN1B)were selected for cytological observation,and the transcriptome was sequenced using the high-throughput Illumine Hiseq2000 sequencing platform,and the sequencing results were then annotated,screened and analysis.The results demonstrated that there were 4284 differentially expressed genes(DEGs)between two lines.GO analysis verified that DEGs were involved in the substance metabolism,cellular processes,transporter protein activities,etc.KEGG analysis indicated that DEGs were enriched in starch and sucrose metabolism,pentose and glucuronide interconversion,glycolysis/gluconeogenesis,and phenyl propane biosynthesis,etc.Transcription factor analysis revealed that DEGs are involved in bHLH,MYB,PHD and other transcription factor families known to be associated with the pollen development.In this work,we observed the anther structure and conducted a transcriptome analysis of alfalfa to provide a reference for studying the mechanism of male sterility in alfalfa.
