摘  要:针对传统消化内科组织粘合剂存在粘合强度低,毒性大的问题,提出一种新型组织粘合剂的制备。试验首先对组合粘合剂的配比进行优化,然后对优化后粘合剂的性能进行研究,结果表明:当明胶质量分数为25%,尿素质量分数为4%,海藻酸钠氧化度为65%,高岭土质量分数为2.5%,微球携带量为2%时制备的粘合剂溶胀率低于300%;固化时间约为20 min,持粘时间约为24 h,粘合强度高于15 kPa;细胞活性值超过80%,满足国际细胞毒性等级I级标准;降解72 h后残余量不超过35%;在8 d时,携带的药物完全释放;对大肠杆菌和金黄色葡萄球菌的抑菌率超过90%,满足生物医用材料抗菌性能的要求。在pH值为6~7,盐浓度0.8%~1%的条件下,粘合剂粘合强度几乎不受影响,即该粘合剂在使用过程中,体液或血液中盐浓度和酸碱度存在一定波动时,表现出良好的适应性。Aiming at the problems of low adhesive strength and high toxicity of traditional tissue adhesive in digestive medicine,a new type of tissue adhesive was prepared.The test first optimized the proportion of the combined adhesive,and then studied the performance of the optimized adhesive.The results show that the swelling rate of the adhesive prepared with 25%gelatin,4%urea,65%sodium alginate oxidation,2.5%kaolin and 2%microsphere carrying capacity is less than 300;when the curing time is about 20 min,and the adhesive holding time is about 24 h,the adhesive strength is higher than 15 kPa;the cell activity value exceeds 80%,meeting the international cytotoxicity grade I standard;after 72 hours of degradation,the residual amount shall not exceed 35%;at 8 d,the drug carried is completely released;the antibacterial rate against Escherichia coli and Staphylococcus aureus is more than 90%,which meets the requirements of antibacterial properties of biomedical materials.Under the conditions of pH value 6~7 and salt concentration 0.8%~1%,the adhesive bonding strength is almost unaffected,that is,the adhesive shows good adaptability when the salt concentration and pH in body fluid or blood fluctuate during use.
