摘 要:目的使用结构光照明显微镜(SIM)揭示3种尿石素亚型对细胞线粒体毒性的差异。方法通过SIM对尿石素孵育后的细胞线粒体超分辨成像,使用图像处理软件Image J进行数据分析。结果SIM在药物浓度10.0μmol/L时即可检测到3种药物亚型的细胞毒性。CCK-8(cell counting kit-8)与SIM在药物浓度40.0μmol/L时均检测到尿石素C的毒性最强。结论尿石素A、B、C 3种亚型中UC的毒性最强。使用超分辨成像技术显微镜在亚细胞水平进行毒性检测,可准确评估和量化低浓度的药物的毒性,促进药物分析和新药研发。Objective To reveal the influences of urolithin subtypes on the distribution of cell mitochondrial morphological subgroups by Structured Illumination Microscopy(SIM).Methods The morphology of mitochondria in cells incubated with urolithins was photographed by SIM,and then the data were analyzed by Image J software Results The cytotoxicity of urolithin A,B,and C at the concentration of 10.0μmol/L were detected.Both CCK-8(Cell Counting Kit 8)and SIM detected the strongest toxicity was from urolithin C at the concentration of 40.0μmol/L.Conclusion Among the three subtypes of urolithin A,B,and C,urolithin C has the strongest toxicity.Using super-resolution microscopy to detect toxicity at the subcellular level can accurately assess and quantify the toxicity of low concentration drugs,promoting drug analysis and new drug development.
