摘  要:目的CyHF-8近红外探针能特异性聚集在棕色脂肪中,现用近红外荧光成像技术,探讨CyHF-8在活体大鼠模型中识别棕色脂肪的应用潜力。方法筛选探针用于大鼠活体荧光成像合适的给药剂量(尾静脉注射0.5、1.0、2.0 mg·kg^(-1));建立糖尿病模型大鼠,经尾静脉注射CyHF-8后,置小动物活体成像仪中进行活体荧光成像。结果尾静脉注射0.5 mg·kg^(-1)CyHF-8适用于SD大鼠活体棕色脂肪的荧光成像;探针聚集于大鼠棕色脂肪中,且荧光信号呈标志性三角形,皮肤和白色脂肪无信号,不干扰成像;与正常组比较,模型组活体棕色脂肪的荧光信号显著下降,与离体脂肪成像结果一致。结论CyHF-8能有效聚集在活体大鼠体的内棕色脂肪中,并释放靶向荧光信号;近红外荧光成像技术可用于无创分析活体模型大鼠内的棕色脂肪。OBJECTIVE The near-infrared probe CyHF-8 specifically accumulates in brown fat,this article intends to explore the application potential of CyHF-8 near-infrared fluorescence imaging technology in identifying brown fat in rats.METHODS Firstly,SD rats were intravenously injected with 0.5,1.0,2.0 mg·kg^(-1) CyHF-8 to screen the appropriate imaging dose;secondly,diabetic rat model was established,which was injected with CyHF-8 via tail vein,and then placed in a small animal in vivo imager for in vivo fluorescence imaging.RESULTS The tail vein injection of 0.5 mg·kg^(-1) CyHF-8 was suitable for fluorescence imaging of brown fat in SD rats;the probe accumulated in the brown fat of rats,and the fluorescence signal showed a signature triangle,while the skin and white fat had no signal,which did not interfere with the imaging;compared with normal rats,the in vivo fluorescence signal of brown fat in the model group was significantly decreased,which was consistent with the results of ex vivo fat imaging.CONCLUSION CyHF-8 effectively aggregates in brown fat in vivo in rats and releases targeted fluorescence signals,NIR fluorescence imaging of CyHF-8 can be used for non-invasive analysis of brown fat in vivo in rats.
