摘 要:目的制备共载胡椒碱与紫杉醇的白蛋白纳米制剂,评价其质量。方法通过正交试验设计,研究制备共载胡椒碱和紫杉醇的白蛋白纳米制剂的最佳处方及实验条件。通过观察白蛋白纳米制剂的外观,测定粒径、电位和稳定性,考察其体外释放的行为,评价白蛋白纳米制剂。结果在优化条件下制备的共载胡椒碱和紫杉醇的白蛋白纳米制剂外观透明乳白色,具有丁达尔效应,粒径为134.8±2.22 nm,PDI 0.214±0.008,Zeta电位为-29.98±2.24 mV,载药量为94.26%±8.52%,包封率为5.66%±0.51%;其在30 d内保存性质稳定;该白蛋白纳米制剂在37℃的PBS缓冲液(pH7.4)中释放较慢,24 h的释放大约为45.3%,48 h释放大约为72.2%。结论制备得到性质优良的共载胡椒碱和紫杉醇的白蛋白纳米制剂,为化疗药物的联合给药及给药方式提供新的参考。OBJECTIVE To prepare piperine paclitaxel and co-loaded albumin nanoparticles and to evaluate their quality.METHODS The best prescription and experimental conditions were investigated for preparing albumin nanoparticles through orthogonal experiment design.The appearance of the albumin nanoparticles was observed,their particle size,potential and stability were measured,and in vitro release behavior was investigated.RESULTS The piperine and paclitaxel co-loaded albumin nanoparticles had a transparent and milky white appearance with the Tyndall effect with the particle size of 134.8±2.22 nm,PDI of 0.214±0.008,Zeta potential of-29.98±2.24 mV,DL of 94.26%±8.52%,and the EE of 5.66%±0.51%.Stability studies showed that the particles were stable within 30 days.The release of albumin nanoparticles was slow in PBS buffer(pH 7.4)at 37℃,where 24 h release was about 45.3%,and 48 h release was approximately 72.2%.CONCLUSION The method in this study can be used to prepare ablumin nanoparticles co-loaded with piperine and paclitaxel,which can provide a new reference for the combination of chemotherapy drugs and the way of administration.
