摘 要:目的探讨苦豆子总碱对大鼠神经行为的改变和氧化损伤的影响。方法将48只SD大鼠随机分为对照组、苦豆子总碱组(低、中、高剂量分别为4、8、16 mg·kg^(-1)·d^(-1)),连续ig给药3周后,进行行为学指标的检测。用ELISA法检测抗氧化指标,观察苦豆子总碱对大鼠学习记忆和氧化损伤的影响。结果在Morris水迷宫实验中,高剂量组大鼠上台潜伏期延长,穿台次数减少;在跳台实验在,高剂量组大鼠通过训练电击次数下降明显;ELISA法结果发现:苦豆子总碱高剂量组能降低血清和脑组织中超氧化物歧化酶、谷胱甘肽过氧化物酶、过氧化物酶的活性,升高丙二醛和脂褐素酶活性。结论苦豆子总碱可导致大鼠学习记忆能力的降低,其机制可能与脑组织中氧自由基清除能力降低及脑组织氧化损伤所致的神经毒性有关。OBJECTIVE To explore the effects of total alkaloids of Sophora alopecuroides L.(TASA)on neurobehavior and oxidative damage in rats.METHODS Forty-eight SD rats,male,were randomly divided into normal control group,TASA 4,8 and 16 mg·kg-1 group after continuous intragastric administration for 3 weeks.Behavioral indexes and antioxidant indexes were detected by ELISA.To observe the effects TASA on learning and memory and oxidative damage in rats.RESULTS In Morris water maze test,on day 5 of training,the stage latency of rats in high-dose group was longer than that in control group,and the number of crossing stage in space cruise test was less than that in control group.The incubation period TASA low-dose and medium-high dose groups was lower than that of the control group.ELISA results showed that the activities of SOD,GSH-Px and POD in serum and brain tissue were decreased,and the contents of MDA and lipofuscin in serum and brain tissue were increased.CONCLUSION The TASA may produce excessive free radicals in brain tissue and affect free radical scavenging,and its mechanism may be related to reducing antioxidant capacity in brain tissue.TASA can reduce learning and memory ability in rats,and oxidative damage of brain tissue may be one of the causes of neurotoxicity and decreased learning and memory ability.
