摘  要:目的优化车前草醇提物有效成分大车前苷和总黄酮的提取工艺,并优化总黄酮的测定方法。方法以大车前苷和总黄酮的提取率作为评价指标,用超声提取法,将提取时间、液料比、超声功率、乙醇浓度、提取次数等作为考察因素,通过正交实验优化提取工艺。结果当超声功率为260 W、提取时间为30 min、液料比为10 mL·g^(-1)、乙醇浓度为50%,提取次数为2次时,大车前苷和总黄酮的提取率分别为6.47±0.21、41.30±1.50 mg·g^(-1)。结论以大车前苷和总黄酮的提取率作为醇提物提取的评价指标,能更好地体现醇提物的特性,可为车前草醇提物的开发和应用提供依据。OBJECTIVE To optimize the extraction process of plantamajoside and total flavonoids from ethanol extracts of Plantaginis Herba,and to optimize the determination method of total flavonoids.METHODS Taking the content of plantamajoside and total flavonoids as the evaluation index,the extraction time,liquid-solid ratio,ultrasonic power,ethanol concentration and extraction times as the influence factors,the extraction method was optimized by orthogonal experiment.RESULTS When the ultrasonic power was 260 W,the extraction time was 30 min,the liquid-material ratio was 10 mL·g^(-1),the ethanol concentration was 50%,and the extraction times were 2 times,the extraction rate of plantamajoside was 6.47±0.21 mg·g^(-1),and the extraction rate of total flavonoids was 41.30±1.50 mg·g^(-1).CONCLUSION In this paper,the extraction rates of plantamajoside and total flavonoids were used as the evaluation indexes of alcohol extract,which could better reflect the characteristics of alcohol extract and provide a basis for the development and application of alcohol extract of Plantaginis Herba.
