摘  要:目的研究山地虎耳草的HPLC指纹图谱,并结合化学计量学分析其药材的质量。方法采用Waters SunFire C18柱(150 mm×4.6 mm,5μm),流动相为乙睛-0.05%磷酸溶液,梯度洗脱,流量1.0 mL·min^(-1),检测波长254 nm,柱温30℃。对不同来源和不同批次的山地虎耳草进行指纹图谱测定,并通过化学计量学系统解决方案软件(ChemPattern 2017)对结果进行处理分析。结果10批次山地虎耳草药材与生成对照指纹图谱的相似度为0.882~0.995,具有较好的一致性。标定了15个共有峰,并对其中4个峰进行了指认,分别为獐牙菜苦苷、当药苷、金丝桃苷、当药醇苷等。主成分分析提取了3个主成分进行综合评价,可以反映样品数据中的绝大部分信息。聚类分析将样品分为2类,10批样品中委托采集的8批样品聚为一类,采集于同一小范围地区(青海贵德县拉脊山)的样品S3、S5聚为一类。结论所用方法科学、准确且简便易行,所建指纹图谱法精密度、稳定性和重复性良好,专属性强,可作为山地虎耳草的质量控制方法。OBJECTIVE To study the HPLC fingerprint of Saxifraga montana,and to analyze the quality of S.montana combined with chemometric analysis.METHODS Waters Sun Fire C18 column(150 mm×4.6 mm,5μm)was used,the mobile phase consisted of acetonitrile-water(containing 0.05% phosphoric acid)with the flow rate of 1.0 mL·min^(-1),the detection wavelength of 254 nm,and column temperature of 30℃.RESULTS The fingerprints of S.montana from different sources and batches were determined,and the results were processed and analyzed by chemometrics system solution software(ChemPattern 2017).The similarity between the fingerprints of 10 batches of S.montana and the generated control was 0.882-0.995,with good consistency.15 common peaks were calibrated and 4 of them were identified-they were swertiamarin,dangyao glycoside,hypericin and dangyao alcohol glycoside.Principal component analysis extracts three principal components for comprehensive evaluation,which reflected most of the information in the sample data.The samples were divided into two categories by cluster analysis.Among the 10 batches of samples,8 batches of samples entrusted for collection were grouped into one class,and the samples S3 and S5 collected from the same small area(Laji mountain,Guide County,Qinghai)were grouped into one class.CONCLUSION The method established in this study is scientific,accurate and simple,and the established fingerprint determination method has good precision,stability,reprodncibility and specificity.It can be used as the quality control method of common“Dida”Tibetan medicine S.montana.
