摘  要:目的:运用Caco-2细胞模型研究白术提取物对人参皂苷Rg1肠吸收的影响。方法:建立Caco-2细胞模型及使用高效液相色谱测定人参皂苷Rg1含量方法,考察在最适细胞生长浓度下,白术提取物对人参皂苷Rg1表观渗透系数(Papp)和转运量的影响。结果:本研究成功构建了Caco-2细胞模型及人参皂苷Rg1的高效液相色谱含量测定方法,研究显示,与正常对照组相比,模型对照组中200μg/mL白术提取物可显著促进400μg/mL人参皂苷Rg1在肠道的吸收,Papp增加超过3倍(P<0.01)。结论:提示白术提取物对人参皂苷Rg1肠吸收有较好的促进作用。Objective:To use Caco-2 cell model to study the effect of ATRACTYLODIS MACROCEPHALAE RHIZOMA extract on the intestinal absorption of ginsenoside Rg1.Methods:This study established the Caco-2 cell model and used high-performance liquid chromatography(HPLC)to determine the content of ginsenoside Rg1.The effect of ATRACTYLODIS MACROCEPHALAE RHIZOMA extract on the apparent permeability coefficient(Papp)and transport amount of ginsenoside Rg1 were investigated under the optimum cell growth concentration.Results:In this study,the Caco-2 cell model and HPLC were successfully established for the determination of ginsenoside Rg1.The study showed that compared with the normal control group,200μg/mL ATRACTYLODIS MACROCEPHALAE RHIZOMA extract significantly promoted the intestinal absorption of 400μg/mL ginsenoside Rg1,and the Papp increased by more than 3 times(P<0.01).Conclusion:ATRACTYLODIS MACROCEPHALAE RHIZOMA extract can promote the intestinal absorption of ginsenoside Rg1,which has certain support for the compatibility theory in traditional Chinese medicine.
