摘  要:为了解猫细小病毒(FPV)在国内的流行状况及其遗传变异和演化方向,本研究对采集的30份疑似感染FPV的肛拭子进行检测,利用常规方法从中分离并鉴定出3株FPV。然后扩增出3株FPV的VP2基因,利用DNAstar等软件将获得的VP2基因与国内外报道的流行株和疫苗株的VP2基因序列进行比对和遗传进化分析。研究结果显示,本文分离的3株FPV均能在猫肾细胞中良好增殖,并产生典型致细胞病变(CPE),在电子显微镜下呈现典型的细小病毒形态特征。序列比对结果显示,此3株FPV的VP2序列与国内外参考毒株的核苷酸和氨基酸序列同源性分别为98.6%~99.8%和98.5%~99.8%;与疫苗株的VP2基因的核苷酸和氨基酸序列同源性分别为99.1%~99.4%和99.1%~99.5%;进化树分析结果显示,本研究分离的FPV与国内流行株及疫苗株均具有较近的遗传距离,并处于同一拓朴群,但是SH612株和ZZ6293株与疫苗株的距离稍微远些,处于不同的拓朴亚群。本文研究结果丰富了中国FPV的流行病学资料,为进一步研发FPV疫苗提供了参考资料。To understand the domestic prevalence of Feline parvovirus(FPV)and its genetic variation and evolution,30 anal swabs suspicious of FPV infection were collected for virus isolation.As a result,3 FPV isolates were obtained and their VP2 gene was sequenced by PCR.Then VP2 gene homology of the epidemic strains and vaccine strains reported in the literature were aligned for genetic evolution analysis.The results showed that the VP2 gene of these 3 isolates had nucleotide and amino acid sequence identities at 98.6%-99.8%and 98.5%-99.8%with domestic/international reference strains and at 99.1%-99.4%and 99.1%-99.5%with the domestic vaccine strain.Furthermore,phylogenic tree analysis showed that these 3 isolates were grouped in the same branch as the domestic epidemic and vaccine strains except that the SH612 and ZZ6293 isolates are not in the same branch as the domestic vaccine strain.These results showed that the domestic FPV was obviously experiencing evolutionary trend,which was the reason why traditional vaccines couldn’t achieve good immune protection.This study enriched the epidemiological data of FPV in China and provided a reference for FPV vaccine research and development.
